Krüppel-like factor (KLF) family members share a three C2H2 zinc finger DNA binding domain, and are involved in cell proliferation and differentiation control in normal as in pathological situations.By loss of heterozygosity (LOH), somatic mutation, or transcriptional suppression by promoter hypermethylation, KLFs can become dysregulated in a variety of malignancies.
At least in part by controlling the expression of cell cycle genes, proteins belonging to the KLF family play a crucial part in the growth and metastasis of various tumor types. KLF4 and KLF6 are generally regarded as tumor suppressor genes, whereas KLF5 encourages cell division. Family members can influence each other's activity through a variety of mechanisms due to their differing transcriptional characteristics. KLF transcription factors most likely create a transcriptional network to regulate cell proliferation because cells can express multiple KLFs. When you take into account the variations in the family members' expression profiles in different cells, the effects of changes in KLF factors can seem contradictory and context-dependent. Finally, KLF variants may interfere with how proteins of the wild type function.
