Telomerase

Telomerase is a DNA polymerase that extends the 3' ends of chromosomes by processively synthesizing multiple telomeric repeats. It is a special ribonucleoprotein (RNP) with a specialized telomerase reverse transcriptase (TERT), telomerase RNA (TER), and other components necessary for TERT's activity (catalytic core), as well as species-specific TER-binding proteins crucial for biogenesis and assembly (core RNP). Other proteins bind telomerase transiently or constitutively to allow association of telomerase and other proteins with telomere.

Telomerase activity is a desirable therapeutic target because it maintains chromosome end structures (telomeres) and cancer cell immortality in the majority of human malignancies. In fact, a telomerase inhibitor is anticipated to have few side effects while still having therapeutic benefits in the majority of cancers. Because they have long telomeres and express telomerase in healthy tissues, adult stem cells should be less affected by telomerase inhibition than cancer cells, which divide quickly and typically have short telomeres.
