Farnesyl transferase is a zinc-dependent enzyme that catalyzes the attachment of a farnesyl lipid group to a cysteine residue of a variety of proteins involved in cell signaling, including the cancer-causing H-Ras protein. Farnesyl transferase controls how the Ras protein is activated, which has an impact on intracellular signaling, cell growth, and proliferation.
An important enzyme in the mevalonate (MVA) pathway is farnesyl transferase. Ras protein, which depends on farnesylation anchoring itself in membrane, is coupled to farnesyl pyrophosphate (FPP), and farnesyl transferase catalyzes the farnesyl moieties of FPP. Small molecules called farnesyl transferase inhibitors target specific intracellular substrate proteins like Ras and prevent them from being farnesylated. A new class of drugs called farnesyl transferase inhibitors targets the signal transduction pathways necessary for the survival and proliferation of various malignant cell types.
