DNA(RNA) Synthesis

RNA synthesis, which is also called DNA transcription, is a highly selective process.Beyond RNA synthesis, RNA polymerase II transcription plays a more active role in the maturation, monitoring, and export of mRNA to the cytoplasm.

In order to fully repair the DNA after a single-strand break, DNA ligase constructs the final phosphodiester bond using the complementary strand of the double helix as a template.Substrates with RNA strands or mismatched base pairs near the ends of the nicked DNA are discriminated against by DNA ligases. Bleomycin (BLM) is a S-independent radiomimetic agent that damages DNA by creating free radicals that attack the C-4′ position in the deoxyribose backbone. Exonuclease/polymerase activity transforms the remaining overhangs into blunt ends. DNA fragments' 3′ ends are adenylated before being ligated with a NEBNext Adaptor with a hairpin loop structure to get the samples ready for hybridization. The two pathways that BET bromodomain inhibition most commonly affects are cell cycle and DNA replication. The translation of mRNA into proteins is inhibited by cycloheximide.
