Arp2 3 Complex

The actin-related proteins Arp2 and Arp3 and the Arp2/3 complex subunits, ARPC1–5, which make up the Arp2/3 complex were first discovered in Acanthamoeba. They are conserved in all eukaryotes, with the exception of some algae, microsporidia, and protists. By virtue of its capacity to produce branched actin filament networks, the complex plays a crucial part in a wide range of cellular processes, including lamellipodia-mediated cell migration, endocytosis, and phagocytosis.

Interaction with actin nucleation-promoting factors (NPFs) is necessary for Arp2/3 activation. Endogenous inhibitory proteins can directly bind to Arp2/3 and compete with NPFs to control its activity, or they can bind to Arp2/3-induced actin filaments and cause branched actin networks to disassemble. Since Arp2/3 inhibition has been linked to reduced cancer progression, neurotoxic side effects from drug abuse, and pathogen invasion of host cells, it has recently attracted more attention.
