Because Wingless is kept in the ER in porcupine mutant Drosophila embryos, it is possible that the porcupine (Porc) protein is involved in secretion or ER transport. Mom-1, a porcupine homolog, serves a similar purpose in C. elegans by encouraging Mom-2's secretion. Porcupine may be involved in the lipid modification of Wnt proteins and shares some homology with a family of o-acyl transferases. On a serine residue in the Wnt protein, a unique type of monounsaturated palmitoylation has been found, and it may also be mediated by pigs.Focal dermal hypoplasia is a genetic disorder that has been linked to the human Porcupine gene. Porcupine, which encodes a multipass transmembrane ER protein, is necessary for Wg to be distributed normally throughout embryos. When expressed in vitro in Drosophila cells, Porc stimulates the processing of Wg and is required for the localization of Drosophila Wnt-3 on the axon tracts of the developing central nervous system.
