Renin

Renin is a monospecific enzyme that catalyzes the rate-limiting step in the synthesis of angiotensin II.Renin and pro-renin bind to the (pro)renin receptor, which results in increased enzymatic activity and additional physiological effects. The renal juxtaglomerular cell is the typical site of renin expression and secretion, and physiological cues tightly control this cell's expression.

In addition to increasing the catalytic conversion of angiotensinogen to angiotensin I (Ang I), prorenin and renin bound to the (pro)renin receptor also upregulate the expression of profibrotic genes. Renin is essential for maintaining electrolyte balance, controlling blood pressure, developing mammalian kidneys, and advancing fibrotic/hypertrophic diseases. An alternative to angiotensin-converting enzyme inhibitors (ACEI) for the treatment of hypertension is renin inhibition.
