PROTACs (Proteolysis Targeting Chimeric Molecules) are heterobifunctional protein degraders and promising targeted therapeutics candidates for cancer.Target protein binders and E3 ligase recruiters, two functional motifs of a PROTAC, are joined by the PROTAC linker.
A PROTAC's linker has a crucial function. The characteristics of the linker (type, length, attachment position) can influence how ligase is formed. The PROTAC:target ternary complex affects the target protein's effective ubiquitination and eventual destruction.
