Molecular Glues

A component of molecular glues is a protein degradation agent based on the ubiquitin-proteasome pathway. Small molecule substances known as "molecular glues" have the ability to stimulate or maintain protein binding. Molecular glue can cause another protein to undergo ubiquitin modification and destruction through the proteasome pathway, which is analogous to PROTAC, if one of the proteins is an ubiquitin ligase. However, in a later classification, these compounds are categorized as functional molecules that act as E3 ligase ligands. Thalidomide, lenalidomide, and pomalidomide, as well as the later-reported CC-90009 and CC-92480, are examples of older medications that fall into this group.
