As a linker molecule, Benzyl-PEG4-MS itself has no intrinsic biological activity; it serves as a structural connector to join a target protein ligand and an E3 ubiquitin ligase ligand in PROTAC synthesis. The mesylate group is an excellent leaving group for nucleophilic substitution (SN2) reactions, allowing the attachment of ligands containing nucleophilic groups such as amines, thiols, or carboxylates. The benzyl group serves as a protecting group for the terminal hydroxyl (which can be deprotected by hydrogenolysis using H2 and Pd/C to reveal a free OH). This free hydroxyl can then be further functionalized (e.g., converted to an amine or tosylate) for additional conjugation.
