Benzyl-PEG5-Ots is a PEG-based PROTAC linker consisting of a benzyl (benzyloxy) group and a tosylate (p-toluenesulfonate, OTs) group connected by a 5-unit PEG spacer (PEG5). This heterobifunctional linker is used in the synthesis of PROTAC molecules, where the tosylate group is a good leaving group that can be displaced by nucleophilic reagents (e.g., thiols, amines) for bioconjugation and PEGylation, and the benzyl group can be removed by hydrogenolysis to reveal a free hydroxyl group for further functionalization. The PEG5 spacer provides water solubility, flexibility, and reduced steric hindrance. This compound is also known as benzyl-pentaethylene glycol tosylate. One ligand is for an E3 ubiquitin ligase, and the other is for the target protein; these two ligands are joined by a linker to form PROTACs. PROTACs target and selectively degrade target proteins by taking advantage of the intracellular ubiquitin-proteasome system.
As a linker molecule, Benzyl-PEG5-Ots itself has no intrinsic biological activity; it serves as a structural connector to join a target protein ligand and an E3 ubiquitin ligase ligand in PROTAC synthesis. The tosylate group is an excellent leaving group for nucleophilic substitution (SN2) reactions, allowing the attachment of ligands containing nucleophilic groups such as amines, thiols, or carboxylates. The benzyl group serves as a protecting group for the terminal hydroxyl (which can be deprotected by hydrogenolysis using H2 and Pd/C to reveal a free OH). This free hydroxyl can then be further functionalized (e.g., converted to an amine or tosylate) for additional conjugation. The PEG5 spacer provides hydrophilicity, flexibility, and reduced steric hindrance, which can facilitate the formation of productive ternary complexes.

No specific in vivo activity has been reported for this linker alone; its activity is derived from the final PROTAC molecule after conjugation with appropriate ligands. The in vivo efficacy of a complete PROTAC is determined in animal models.
