NF-κB-IN-9 (TR2; 1 nM–10 μM) has an EC50 value of 68.9 nm[1], which reduces NO generation in RAW 264.7 macrophages. Using 10 μM of NF-κB-IN-9 and 30 seconds of sonication, MCF-7 cell growth was suppressed and apoptosis was induced. When sonicated for 30 seconds, 2 minutes, and 5 minutes, respectively, NF-κB-IN-9 demonstrates sonocytotoxicity to MCF-7 cells, with IC50 values of 1.6 μM, 0.55 μM, and 0.29 μM [1].

In a xenograft mouse model, NF-κB-IN-9 (TR2; 1.5 mg/kg; i.v.; days 0 and 10, respectively) demonstrated good tolerance and antitumor activity. The brain exhibits NF-κB-IN-9 fluorescence, suggesting that TR2 can cross the blood-brain barrier [1].
