Selectivity for the 5-HT1A receptor over other biogenic amine receptors is exhibited by U92016A (U-92016A). In Chinese hamster ovary cells expressing the human 5HT1A receptor, U92016A has an intrinsic activity of 0.82 relative to 5-HT and reduces the Forskolin-induced increase in cyclic AMP synthesis[2].

In mice, U92016A (U-92016A) potently lowers rectal temperature. In rats, U92016A also causes the 5-HT-mediated syndrome and reduces 5-hydroxytryptophan accumulation in a dose-dependent manner. In addition, U92016A lowers arterial blood pressure in rats that develop spontaneous hypertension and suppresses sympathetic nerve activity in cats. Excellent potency and a prolonged duration of action are displayed by U92016A. In addition, U92016A is active in two assays measuring social interaction and suppresses the firing of dorsal raphe 5-HT neurons. U92016A has a po bioavailability of 45%[2].
