Product Description
Adipic acid-13C is 13C (carbon 13) labelled Adipic acid. Adipic acid has been found to be associated with HMG-CoA lyase deficiency, carnitine kinase deficiency, and medium-chain acyl dehydrogenase deficiency, all of which are caused by innate metabolic abnormalities.
Adipic acid-13C (Hexanedioic acid-13C) is the 13C-labeled form of adipic acid. It has a molecular formula of C5^{13}CH10O4 and a molecular weight of 147.13 g/mol. Adipic acid is an orally active compound with anti-HSV-1 activity. It has low toxicity and can be used as a food additive and a gelling agent.
Biological Activity I Assay Protocols (From Reference)
Targets
Adipic acid-13C does not have a specific pharmacological receptor target. As a stable isotope-labeled compound, it is used as a tracer in metabolic studies. Adipic acid itself is an endogenous metabolite associated with various inborn errors of metabolism. The 13C labeling allows for the tracking of its metabolic fate in biological systems.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [1].
In vitro, Adipic acid-13C is not used for conventional biological activity assays. Its primary utility is as a tracer in metabolic studies. Stable heavy isotopes of hydrogen, carbon, and other elements have been incorporated into drug molecules as tracers for quantitation during the drug development process. Adipic acid, the unlabeled compound, has anti-HSV-1 activity.
ln Vivo
In vivo, Adipic acid-13C is used as a metabolic tracer to study fatty acid metabolism and to diagnose inborn errors of metabolism. By administering the labeled compound and measuring the incorporation of 13C into downstream metabolites using mass spectrometry, researchers can map metabolic pathways. It is not used for therapeutic purposes.
Enzyme Assay
For non-cell-based assays, Adipic acid-13C is used as an analytical standard. Its identity and purity are confirmed using techniques such as NMR, HPLC, and mass spectrometry. It is not used in receptor binding assays. Its role is to serve as a quantitative tracer in metabolic studies, enabling the tracking of carbon atoms through metabolic pathways.
Cell Assay
For in vitro cellular assays, cells are cultured in media containing Adipic acid-13C. The compound is taken up by cells and metabolized through various pathways. After a defined incubation period, cells are harvested, and metabolites are extracted. The incorporation of 13C into metabolites is analyzed using LC-MS/MS or GC-MS to determine metabolic fluxes and pathway activities.
Animal Protocol
For in vivo animal studies, Adipic acid-13C can be administered to rodents via oral gavage. Blood or tissue samples are collected at various time points post-administration. The samples are processed and analyzed by mass spectrometry to track the distribution and metabolism of the labeled compound. This allows for the study of fatty acid metabolism and related metabolic disorders.
ADME/Pharmacokinetics
Adipic acid-13C has a molecular weight of 147.13 g/mol and a molecular formula of C5^{13}CH10O4. It is soluble in DMSO at 100 mg/mL (679.67 mM). The compound should be stored at -20°C for up to 3 years or at 4°C for up to 2 years. In solvent, it can be stored at -80°C for 6 months or at -20°C for 1 month. It is for research use only and is not intended for human consumption.
Toxicity/Toxicokinetics
The toxicity profile of Adipic acid-13C is not applicable, as it is used as a metabolic tracer at very low concentrations. Adipic acid itself has low toxicity. The 13C labeling is not expected to introduce new toxicities, as carbon-13 is a stable, non-radioactive isotope. The compound is not intended for human consumption. Standard safety precautions should be observed when handling the compound.
