NH4-6 can efficiently and selectively inhibit the activity of purified SIRT2 deacetylase with an IC50 value of 0.032 μM. It has weak activity against SIRT1 and SIRT6 and does not inhibit SIRT3 at a concentration of 83 μM[1]. In MCF7 and MDA-MB-231 breast cancer cells, NH4-6 (25-50 μM; permeability assay 6 h) is less cytotoxic than TM (25 μM), but stronger at a concentration of 50 μM, which is related to the concentration-dependent cellular uptake caused by its excellent water solubility[1]. In MCF7 breast cancer cells, NH4-6 (12 μM; 10 days) has a stronger inhibitory effect on non-anchorage-dependent growth than TM (12 μM)[1]. NH4-6 (25–100 μM; 6 h) increased the acetylation level of α-tubulin in MCF7 breast cancer cells (a marker of SIRT2 inhibition) at concentrations of 50 μM and 100 μM, but had no effect at a concentration of 25 μM, consistent with its concentration-dependent cellular uptake [1]. NH4-6 (10–100 μM) inhibited SIRT1 activity in MCF7 breast cancer cells only at a concentration of 100 μM (as determined by p53 acetylation levels), which may be due to its increased solubility, allowing it to accumulate sufficiently within cells [1].
