ACTH 1-39 is non-toxic to neurons in the concentration range of 100-400 nM, but it protects neurons from excitotoxic neuronal death induced by glutamate (100 μM), NMDA (1 mM), AMPA (50 μM), and phycocyanine (25 μM). 400 nM ACTH provides significant protection in all of these cases. Both 200 nM and 400 nM ACTH protect neurons from damage caused by quinolinic acid (25 μM). ACTH also protects neurons from 2 μM H₂O₂-induced cell death (H₂O₂ produces reactive oxygen species (ROS)), with 400 nM ACTH showing significantly stronger protection than 200 nM. ACTH provides some protection against rapidly released nitric oxide (NO) from NOC-12, but not against slowly released nitric oxide from NOC-18. ACTH (200 or 400 nM) can protect neurons from the cytotoxic effects of asteroids (10-20 nM), a classic drug that induces cell death through apoptosis. ACTH can reduce the cell death rate from 80% to 55%[1].

Compared with the saline/IgG group, intraventricular injection of ACTH significantly reduced cumulative food intake during the observation period. Injection of ACTH antibody into the paraventricular nucleus (PVN) eliminated the anorexia of ACTH. In rats injected with α-MSH antibody into the PVN and ACTH into the ventricle, intraventricular infusion of ACTH significantly reduced cumulative food intake, which was as low as that of the intraventricular ACTH injection and PVN IgG injection groups. Compared with the IgG treatment group, injection of ACTH antibody or α-MSH antibody into the PVN significantly increased cumulative food intake; the combined use of the two antibodies did not further increase food intake [2].
