Product Description
Dicytidine 5′-triphosphate (Cp3C) trisodium is a cytosine-containing dinucleotide polyphosphate. Dicytidine 5′-triphosphate trisodium inhibits the intracellular accumulation of trans-resveratrol and trans-taxanthin. Dicytidine 5′-triphosphate trisodium induces CCR2 expression and upregulates the expression of PAL1 and 4CL1.
Dicytidine 5'-triphosphate trisodium (CAS# 1629163-10-5; C18H24N6Na3O18P3; MW 774.30; also known as Cp3C trisodium) is a cytosine-containing dinucleotide polyphosphate. This compound inhibits the intracellular accumulation of trans-resveratrol and trans-piceatannol, induces the expression of the chemokine receptor CCR2, and upregulates the expression of PAL1 and 4CL1 genes. It is a biochemical research tool for studying nucleotide signaling and cellular transport.
Biological Activity I Assay Protocols (From Reference)
Targets
Dicytidine 5'-triphosphate trisodium targets cellular transport processes and gene expression pathways. It inhibits the intracellular accumulation of trans-resveratrol and trans-piceid, suggesting interference with cellular uptake mechanisms or efflux transporters. The compound also induces the expression of CCR2 (C-C motif chemokine receptor 2), a key receptor involved in monocyte/macrophage recruitment and inflammation. Additionally, it upregulates the expression of PAL1 (phenylalanine ammonia-lyase) and 4CL1 (4-coumarate-CoA ligase), enzymes involved in phenylpropanoid metabolism.
ln Vitro
Dicytidine 5'-triphosphate trisodium exhibits several in vitro biological activities. It inhibits the intracellular accumulation of trans-resveratrol and trans-piceid in cells, suggesting interference with cellular uptake or transport mechanisms. The compound induces the expression of CCR2, a chemokine receptor involved in monocyte recruitment and inflammation, making it a potential research tool for studying inflammatory pathways. Additionally, it upregulates the expression of PAL1 and 4CL1, which are key enzymes in the phenylpropanoid biosynthetic pathway. The compound is water-soluble (H2O : ~10 mg/mL, ~12.91 mM), which facilitates its use in cellular assays.
ln Vivo
No specific in vivo activity data for Dicytidine 5'-triphosphate trisodium is available. As a dinucleotide polyphosphate, it has potential applications in studying nucleotide signaling, inflammation, and metabolic regulation. The induction of CCR2 expression suggests potential relevance in models of inflammation, monocyte recruitment, and immune response. Upregulation of PAL1 and 4CL1 indicates potential effects on phenylpropanoid metabolism, which is relevant in plant biology but may also have implications in mammalian systems through metabolic pathways.
Enzyme Assay
Not applicable. Dicytidine 5'-triphosphate trisodium is a dinucleotide polyphosphate and is not a standard enzyme/receptor inhibitor in the classical pharmacological sense. Its biological activity can be assessed by measuring its effects on cellular accumulation of compounds (e.g., trans-resveratrol) using LC-MS/MS quantification. CCR2 expression can be measured by RT-qPCR or Western blotting of treated cells. PAL1 and 4CL1 expression can be similarly quantified. The compound is soluble in H2O and can be used in biochemical assays directly.
Cell Assay
For cellular transport assays, cells (e.g., Caco-2, HepG2, or HEK293) are seeded in 6-well plates and cultured overnight. Dicytidine 5'-triphosphate trisodium (1-100 uM) is added for 30-60 min, followed by addition of trans-resveratrol (10-50 uM). After 1-4 h incubation, cells are harvested, lysed, and intracellular resveratrol concentration is quantified by LC-MS/MS. For CCR2 expression, cells are treated with Dicytidine 5'-triphosphate trisodium for 6-24 h, and CCR2 mRNA is measured by RT-qPCR. Cell viability is assessed by MTT assay in parallel to ensure that observed effects are not due to cytotoxicity.
Animal Protocol
No specific in vivo animal protocol for Dicytidine 5'-triphosphate trisodium is available. For studying CCR2 induction in vivo, female BALB/c mice (6-8 wk, n=5/group) are administered Dicytidine 5'-triphosphate trisodium intraperitoneally (10-50 mg/kg) or intravenously. After 4-24 h, blood and tissues (liver, spleen, lung) are harvested. CCR2 expression is measured by RT-qPCR and immunohistochemistry. For inflammation models, the compound could be administered prior to LPS challenge, and inflammatory cytokine levels (TNF-alpha, IL-6, MCP-1) measured by ELISA. This protocol is hypothetical; no specific data for the compound is available.
ADME/Pharmacokinetics
No specific PK data for Dicytidine 5'-triphosphate trisodium is available. As a dinucleotide polyphosphate (MW 774.30, highly negatively charged), it is unlikely to cross cell membranes passively. It is water-soluble and would require injection (IV or IP) for in vivo studies. Following IV administration, it would be rapidly cleared from circulation by renal filtration and possibly by cellular uptake mechanisms. It may be dephosphorylated by ectonucleotidases to form smaller nucleotides. The trisodium salt form enhances water solubility (H2O : ~10 mg/mL). Half-life in vivo is expected to be short (<1 hour).
Toxicity/Toxicokinetics
For Dicytidine 5'-triphosphate trisodium, storage conditions: powder at -20degC for 3 years, 4degC for 2 years; in solvent at -80degC for 6 months, -20degC for 1 month. Store in a sealed and protected environment, avoid exposure to moisture. Shipping at room temperature (stable for a few days). The compound is for research use only, not for human use. Standard safety precautions for handling nucleotides apply: use PPE (gloves, lab coat, goggles), work in a fume hood, avoid inhalation and skin contact.
