Biological Activity I Assay Protocols (From Reference)
Targets
NCT-14b targets histone deacetylase 6 (HDAC6), a class IIb histone deacetylase that deacetylates α-tubulin, cortactin, and other non-histone substrates. HDAC6 is involved in various cellular processes including cell motility, protein degradation, and aggresome formation. By selectively inhibiting HDAC6, NCT-14b increases acetylation of α-tubulin and other HDAC6 substrates, affecting microtubule dynamics and cellular signaling. The compound shows high selectivity for HDAC6 over other HDAC isoforms.
ln Vitro
In vitro, NCT-14b potently inhibits HDAC6 with an IC₅₀ of 84 nM. It shows >40-fold selectivity over other HDAC isoforms (IC₅₀ >3.5 μM). The compound inhibits cell growth of estrogen receptor α-positive MCF-7 breast cancer cells treated with estrogen but not untreated cells. This suggests estrogen-dependent activity. Detailed in vitro data are available in the primary literature. The compound is a valuable tool for studying HDAC6-selective inhibition.
ln Vivo
In vivo data for NCT-14b are limited in publicly available sources. Enzyme Assay
In vitro HDAC activity assays for NCT-14b typically use recombinant HDAC6 enzyme or HeLa nuclear extracts with fluorogenic substrate (e.g., Boc-Lys(Ac)-AMC). The compound is dissolved in DMSO and diluted in assay buffer (50 mM Tris-HCl, pH 8.0, 137 mM NaCl, 2.7 mM KCl, 1 mM MgCl₂). Cell Assay
For cell-based assays, MCF-7 breast cancer cells are cultured in DMEM with 10% FBS and estrogen supplementation. Cells are seeded in 96-well plates and treated with NCT-14b at various concentrations (0.01-10 μM) for 24-72 hours. Cell viability is assessed by MTT or CellTiter-Glo. α-Tubulin acetylation is measured by Western blotting to confirm HDAC6 inhibition. Cell cycle analysis is performed by flow cytometry. Gene expression changes are analyzed by RT-PCR. All treatments include vehicle controls and are performed in triplicate.
Animal Protocol
In vivo, NCT-14b may be administered to rodents via intraperitoneal (IP) or intravenous (IV) injection. The compound is formulated in a suitable vehicle such as DMSO, PEG, or saline-based solutions. For tumor xenograft models, immunodeficient mice bearing MCF-7 tumors are dosed at various regimens. Endpoints include tumor volume measurement, survival analysis, and biomarker assessment (α-tubulin acetylation in tumor tissues). Blood and tissue samples are collected for pharmacokinetic and pharmacodynamic analysis. All procedures follow institutional animal care guidelines.
