Endogenous metabolite; influenza virus neuraminidase (EC 3.2.1.18); sialic acid-binding immunoglobulin-like lectins (Siglecs); selectins. Neu5Ac acts as both a substrate and competitive inhibitor of viral neuraminidases, blocking viral budding. It also modulates leukocyte extravasation through selectin interactions and serves as a ligand for Siglec receptors on immune cells.

| ln Vitro |
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [1].
As a labeled metabolite, Neu5Ac-13C itself exhibits the same intrinsic biological activity as unlabeled Neu5Ac. It competitively inhibits Clostridium perfringens sialidase with Ki in the micromolar range. In influenza neuraminidase inhibition assays, Neu5Ac derivatives typically show IC50 values from low micromolar to sub-micromolar. It also scavenges hydrogen peroxide, contributing to cellular antioxidant defense. |
| ln Vivo |
In vivo, Neu5Ac-13C is used for metabolic tracing of sialylation pathways in animal models. Following administration, the 13C label is incorporated into tissue glycoproteins and gangliosides, enabling quantification of sialic acid turnover in brain, liver, and serum. Neu5Ac supplementation has shown neuroprotective effects in rodent models and modulates immune responses by altering sialylation patterns on cell surfaces.

| Enzyme Assay |
Neuraminidase inhibition is measured using 4-methylumbelliferyl-N-acetyl-α-D-neuraminic acid (4-MU-NANA) as substrate. Enzyme (0.01 μM) is pre-incubated with varying concentrations of Neu5Ac-13C in assay buffer at room temperature for 10-60 minutes. Substrate is added to a final concentration of 1 mM, and fluorescence (Ex 365 nm/Em 450 nm) is monitored every minute for 30 minutes. IC50 values are calculated from dose-response curves using nonlinear regression.
|
| Cell Assay |
For metabolic labeling, cells are cultured in medium supplemented with 100-500 μM Neu5Ac-13C for 24-72 hours. Cells are harvested, and glycoproteins are extracted. Incorporated 13C-labeled sialic acid residues are detected by LC-MS/MS after acid hydrolysis and DMB derivatization. For viral infection studies, cells are pre-treated with Neu5Ac-13C, infected with influenza virus, and viral titers are measured by plaque assay.
|
| Animal Protocol |
For in vivo metabolic tracing, mice receive Neu5Ac-13C (50-200 mg/kg) via oral gavage or intraperitoneal injection. Tissues (brain, liver, serum) are collected at defined time points (1-48 hours). Sialic acid is released by acid hydrolysis, purified, and analyzed by LC-MS/MS to quantify 13C enrichment. For neuroprotection studies, rodents receive Neu5Ac-13C daily for 1-2 weeks, followed by behavioral testing and histological analysis.

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations
Oral Formulation 1: Suspend in 0.5% CMC Na (carboxymethylcellulose sodium) Oral Formulation 2: Suspend in 0.5% Carboxymethyl cellulose Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
