Product Description
Cholesterol-d6 is the deuterium labelled form of Cholesterol. Cholesterol is a major sterol in mammals, accounting for 20-25% of the structural components of the plasma membrane. The plasma membrane is highly permeable/penetrable to water but relatively impermeable/penetrable to ions and protons. Cholesterol plays important roles in determining the fluidity and permeability characteristics of membranes and the function of transport and signaling proteins. Cholesterol is also an endogenous estrogen-related receptor alpha (ERRα) agonist.
Cholesterol-d6 (CAS 60816-17-3) is the deuterium-labeled form of Cholesterol, the major sterol in mammals. Cholesterol makes up 20-25% of the structural component of the plasma membrane. Cholesterol-d6 features six deuterium atoms at the 26,26,26,27,27,27 positions. It is intended for use as an internal standard for the quantification of cholesterol by GC- or LC-MS. The compound has a molecular formula of C27H40D6O and a molecular weight of 392.69.
Biological Activity I Assay Protocols (From Reference)
Targets
Cholesterol-d6 targets the same biological pathways as unlabeled cholesterol. Cholesterol is a major sterol produced in mammalian cells that is required for cell viability and proliferation. It is a structural component of plasma membranes and serves as a precursor for steroid hormones and bile acids. The deuterated form is used to study lipid metabolism, cardiovascular health, and steroid biosynthesis.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [1].
In vitro, Cholesterol-d6 is used as a tracer to study cholesterol metabolism, membrane dynamics, and lipid signaling. The compound is added to cell culture media to track cholesterol uptake, incorporation into membranes, and metabolic fate. It does not exhibit intrinsic pharmacological activity but serves as a quantitative tool for studying cellular lipid metabolism.
ln Vivo
In vivo, Cholesterol-d6 is used in metabolic studies to trace cholesterol absorption, distribution, metabolism, and excretion. The deuterium label allows for precise quantification of the compound and its metabolites in biological fluids and tissues using mass spectrometry. It is used in research on lipid metabolism and cardiovascular health.
Enzyme Assay
As an analytical standard, Cholesterol-d6 is used in lipidomics and sterol analysis. Typical protocols involve spiking the labeled compound into samples prior to extraction and analysis by GC-MS or LC-MS. The compound serves as an internal standard to correct for matrix effects and instrument variability.
Cell Assay
In vitro cell culture experiments using Cholesterol-d6 involve treating cells with the labeled sterol and analyzing its uptake and metabolism by mass spectrometry. The compound can be used to study cholesterol metabolism, membrane dynamics, and the effects of cholesterol on cellular function in various cell types.
Animal Protocol
In vivo animal studies using Cholesterol-d6 involve administering the compound to rodents via oral gavage. Blood samples are collected at various time points, and the concentration of the labeled compound is measured by mass spectrometry. Tissue distribution and metabolic profiling can also be performed.
ADME/Pharmacokinetics
Cholesterol-d6 exhibits pharmacokinetic properties similar to those of unlabeled cholesterol. Cholesterol is absorbed from the gastrointestinal tract, incorporated into lipoproteins, and distributed throughout the body. It is metabolized in the liver and excreted primarily in bile. The deuterium label provides a distinct mass shift for analytical detection.
Toxicity/Toxicokinetics
Cholesterol-d6 is considered safe for research use at typical concentrations. Cholesterol is a naturally occurring sterol with a well-established safety profile at physiological concentrations. As a stable isotope-labeled compound, it is not intended for therapeutic use and is handled under standard laboratory safety practices.
