Description: Bemegride (3-Ethyl-3-methylglutarimide; Antibarbi; Agipnon; Ahypnon; Eukraton; Malysol; Mikedimide; Zentraleptin) is apotent central nervous system stimulant and effective antidote for barbiturate poisoning. Ethinamate is a short-acting carbamate-derivative, sedative-hypnotic medication used to treat insomnia. Regular use leads to tolerance, and it is usually not effective for more than 7 days. Structurally, it does not resemble the barbituates, but it shares many effects with this class of drugs. The depressant effects of ethinamate are, however, generally milder than those of most barbiturates.

    In Vitro

In vitro activity: Bemegride has an antagonistic action on the GABAA receptor, suppressing both GABA- and pentobarbitone-evoked whole-cell currents to similar extents.

In VivoLong-term oral administration to the rat of barbitone, alone or together with the analeptics bemegride or pentylenetetrazol, show that the intensity of the withdrawal syndrome generally parallels the degree of associated CNS depression.
