Product Description
8-Bromo-2'-deoxyadenosine is an analogue of purine nucleoside. Purine nucleoside analogues have broad-spectrum anticancer effect targeting indolent lymphoid malignancies. The anti-cancer mechanism in this process relies on inhibiting DNA synthesis and inducing apoptosis.
8-Bromo-2'-deoxyadenosine (8-BrdA) is a modified 2'-deoxyadenosine having a bromo substituent at position 8 of the adenosine ring system. It is a purine nucleoside analog that exerts anticancer effects primarily through the inhibition of DNA synthesis and the induction of apoptosis. It has demonstrated broad antitumor activity, particularly against indolent lymphoid malignancies. It is used to measure the kinetics of oxidative DNA damage and to study glycosidic bond cleavage reactions. It is a research-grade compound not approved for human therapeutic use.
Biological Activity I Assay Protocols (From Reference)
Targets
Purine nucleoside metabolism pathways; DNA synthesis. As a purine nucleoside analog, 8-Bromo-2'-deoxyadenosine is incorporated into DNA, disrupting nucleic acid synthesis. The bromine atom at position 8 may affect DNA structure and stability. The compound targets rapidly dividing cells, particularly lymphoid malignancies.
ln Vitro
8-Bromo-2'-deoxyadenosine demonstrates activity as a purine nucleoside analog in cellular assays. It exerts anticancer effects primarily through the inhibition of DNA synthesis and the induction of apoptosis. It has demonstrated broad antitumor activity, particularly against indolent lymphoid malignancies. Detailed IC50 values have not been disclosed.
ln Vivo
8-Bromo-2'-deoxyadenosine has potential applications in cancer research due to its ability to interfere with DNA synthesis in rapidly dividing cells, leading to cell death or growth inhibition. It is used to measure the kinetics of oxidative DNA damage and to study glycosidic bond cleavage reactions. In vivo studies would be needed to evaluate its therapeutic potential.
Enzyme Assay
In vitro assays for purine nucleoside analogs typically involve measuring inhibition of DNA synthesis using radiolabeled thymidine incorporation. For DNA damage studies, the compound is used to measure oxidative DNA damage kinetics and glycosidic bond cleavage reactions. Cell viability is assessed by MTT or CellTiter-Glo assays.
Cell Assay
Cells (e.g., lymphoid malignancy cell lines or other cancer cell lines) are cultured and treated with 8-Bromo-2'-deoxyadenosine at concentrations typically ranging from 0.1–100 μM for 24–72 hours. DNA synthesis inhibition is measured by BrdU incorporation. Cell viability is assessed by standard assays. DNA damage is evaluated by comet assay or γ-H2AX staining.
Animal Protocol
In vivo animal studies for 8-Bromo-2'-deoxyadenosine have not been reported. The compound is primarily used as a research tool for DNA damage and nucleoside analog studies. Future studies in xenograft models would be needed to evaluate in vivo efficacy.
