| Targets |
4-iodo-SAHA targets class I and class II histone deacetylases (HDACs). It binds the zinc ion in the catalytic site of HDACs and inhibits their deacetylase activity. This leads to increased histone acetylation, chromatin remodeling, and re-expression of silenced tumor suppressor genes. It induces apoptosis and inhibits cell proliferation. It also suppresses HDAC7.

| ln Vitro |
4-Iodo-SAHA (0.1-100 μM; 48 h) suppresses the HT29, JA16, HL60, U937, and Skbr3 cell lines[1].
4-Iodo-SAHA (2 μM; 6-24 h) modifies the levels of p21 and acetylated H4 in SKBR3 cells[1]. In vitro, 4-iodo-SAHA inhibits class I and class II HDACs with EC50 values of 1.1, 0.95, 0.12, 0.24, and 0.85 µM for Skbr3, HT29, U937, JA16, and HL60 cells, respectively. It is 10-fold more potent than SAHA in inhibiting U937 cell proliferation. It induces apoptosis and elevates histone acetylation. It suppresses HDAC7 and resensitizes drug-resistant cells. |
| ln Vivo |
4-Iodo-SAHA (1k) (50 mg/kg; p.o. five times weekly for two weeks) exhibits comparable toxicity to both SAHA-treated and control mice[1].
In vivo, 4-iodo-SAHA is orally active and has antiproliferative activity in various cancer cell lines. It is a promising monotherapy candidate warranting further investigation in advanced disease settings. It has not been approved for therapeutic use. |
| Enzyme Assay |
For non-cell enzyme assays, recombinant HDACs are incubated with 4-iodo-SAHA and a fluorogenic acetyl-lysine substrate. Deacetylase activity is measured by fluorescence release. IC50 values are calculated from dose-response curves. Selectivity is assessed against different HDAC isoforms.

| Cell Assay |
Cell Line: SKBR3, HT29, U937, JA16 and HL60 cell lines
Concentration: 0.1-100 μM Incubation Time: 48 h Result: Inhibited SKBR3, HT29, U937, JA16 and HL60 cell lines with EC50s of 1.1, 0.95, 0.12, 0.24, 0.85 and 1.3 μM , respectively. Showed 10-fold potent as an inhibitor of U937 cell line compared to SAHA. In cell assays, cancer cell lines (e.g., U937, SKBR3, HT29) are treated with serial dilutions of 4-iodo-SAHA. Cell viability is measured by MTT or SRB. Apoptosis is assessed by Annexin V staining or caspase-3 activity. Histone acetylation is measured by Western blot. HDAC7 expression is measured by Western blot. |
| Animal Protocol |
8-week-old fvb mice
50 mg/kg Oral gavage; 50 mg/kg five times per week; for 2 weeks In vivo animal protocols use mice bearing tumor xenografts. 4-iodo-SAHA is dissolved in DMSO/PEG/saline and administered orally at doses of 10-100 mg/kg. Tumor volume is measured twice weekly. Tumor tissue is analyzed for histone acetylation and apoptosis markers. Pharmacokinetic parameters are determined by LC-MS. |

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations
Oral Formulation 1: Suspend in 0.5% CMC Na (carboxymethylcellulose sodium) Oral Formulation 2: Suspend in 0.5% Carboxymethyl cellulose Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals). *Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
