Product Description
Uridine-13C-3 is 13C (carbon 13)-labeled Uridine. Uridine (β-Uridine) is a nucleoside compound consisting of uracil and ribose rings, which are connected by β-N1-glycoside bonds.
Uridine 13C-3 (CAS#: 478511-16-9) is a stable isotope-labeled form of uridine, a nucleoside and one of the four basic components of RNA. The 13C label is incorporated at the 5'-position of the ribose moiety, enabling tracing of RNA synthesis, nucleotide metabolism, and cellular dynamics. It is primarily used in metabolic studies and tracer experiments to investigate nucleoside metabolism.
Biological Activity I Assay Protocols (From Reference)
Targets
Endogenous metabolite; uridine phosphorylase; uridine kinase; RNA polymerase; pyrimidine nucleoside transporters. Uridine is phosphorylated to UMP by uridine kinase and converted to UDP and UTP for RNA synthesis. It also serves as a substrate for uridine phosphorylase, yielding uracil and ribose-1-phosphate for salvage pathways.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may have an effect on a drug's pharmacokinetics and metabolic properties, it is a cause for concern [1].
In vitro, Uridine 13C-3 is used for pulse-labeling of nascent RNA in cultured cells. It is incorporated into RNA within 15-30 minutes of addition to culture medium. The compound enables quantification of RNA synthesis rates and half-life by tracking 13C enrichment in extracted RNA. It also supports nucleotide salvage pathway studies.
ln Vivo
In vivo, Uridine 13C-3 is used for metabolic tracing of RNA turnover in animal tissues. Following administration, the 13C label is incorporated into tissue RNA, enabling quantification of RNA synthesis and degradation rates in liver, brain, and other organs. It also supports studies of nucleoside transport and pyrimidine metabolism in vivo.
Enzyme Assay
For RNA synthesis assays, cells are pulsed with Uridine 13C-3 (100-500 μM) for 15-60 minutes. RNA is extracted using TRIzol or column-based methods. 13C enrichment in RNA is quantified by LC-MS/MS after enzymatic digestion to nucleosides. RNA half-life is determined by chase experiments with excess unlabeled uridine.
Cell Assay
For pulse-chase labeling, cells are cultured in medium containing Uridine 13C-3 (100-500 μM) for 15-60 minutes (pulse), then washed and cultured in medium with excess unlabeled uridine (3 mM) for 0-8 hours (chase). RNA is extracted at each time point, and 13C enrichment is quantified by LC-MS/MS to determine RNA half-life.
Animal Protocol
For in vivo RNA turnover studies, mice receive Uridine 13C-3 (50-200 mg/kg) via intraperitoneal injection or oral gavage. Tissues (liver, brain) are collected at defined time points (0-24 hours). RNA is extracted, digested to nucleosides, and analyzed by LC-MS/MS to quantify 13C enrichment. For kinetic studies, multiple doses or continuous infusion may be used.

Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 3.6666 mL 18.3332 mL 36.6663 mL
5 mM 0.7333 mL 3.6666 mL 7.3333 mL
10 mM 0.3667 mL 1.8333 mL 3.6666 mL
