Umbralisib tosylate (TGR-1202) is an orally bioactive dual inhibitor of PI3Kδ and casein kinase-1-ε (CK1ε) with anticancer and immunomodulatory effects. It inhibits PI3Kδ and casein kinase-1-ε (CK1ε) with EC50 of 22.2 nM and 6.0 μM, respectively.

Human whole blood CD19 cell proliferation is half-maximally inhibited by umbralisib tosylate between 100 and 300 nM[3]. In human lymphoma and leukemia cell lines, umbralisib tosylate (10 nM-100 μM) suppresses phosphorylated AKT at Ser473 in a concentration-dependent manner[4]. In the DLBCL cell line LY7, umbralisib tosylate (15–50 μM) potently suppresses the expression of c-Myc. Its distinct structural characteristics make it appropriate for targeting CK1ε in lymphoma cells.

Using the MOLT-4 cell line, umbralisib tosylate (150 mg/kg, po daily) dramatically reduces the tumors by day 25 in a subcutaneous xenograft model of T-cell acute lymphoblastic leukemia (T-ALL) in NOD/SCID mice[4].
