Product Description
Tiopronin-13C,d3 is a deuterium labelled form ofTiopronin.
Tiopronin 13C D3 (CAS 1189695-13-3) is the 13C- and tri-deuterated form of tiopronin (N-(2-mercaptopropionyl)glycine), a thiol-containing pharmaceutical agent used for treating cystinuria and as an antidote to heavy metal poisoning. The stable isotope labels (13C and D3) are incorporated to create a stable isotope-labeled internal standard for LC-MS/MS quantification. Tiopronin is a diffusible antioxidant that reduces disulfide bonds in cystine.
Biological Activity I Assay Protocols (From Reference)
Targets
Tiopronin acts as a thiol-containing antioxidant and reducing agent. It reduces cystine to cysteine, preventing cystine stone formation in the urinary tract. It also serves as an antidote for heavy metal poisoning by chelating metals.
ln Vitro
Tiopronin 13C D3 is a stable isotope-labeled analog. The parent compound tiopronin is a diffusible antioxidant with reducing activity. It reduces disulfide bonds and serves as a free radical scavenger. It has been used clinically for cystinuria and heavy metal poisoning.
ln Vivo
Tiopronin 13C D3 is primarily used as an analytical reference standard. The parent compound tiopronin has been evaluated in clinical trials for cystinuria. It is well absorbed orally and excreted renally.
Enzyme Assay
The thiol quantification assay measures the reducing activity of tiopronin using Ellman's reagent (DTNB). Tiopronin reduces DTNB to TNB, which is quantified spectrophotometrically at 412 nm. The amount of reduced thiol is calculated from a standard curve.
Cell Assay
Cell-based assays for antioxidant activity use cells exposed to oxidative stress (e.g., H2O2 or heavy metals). Cells are treated with serially diluted tiopronin, and cell viability is assessed by MTT or ATP-based assays. Intracellular glutathione levels and reactive oxygen species are measured by fluorescent probes.
Animal Protocol
In vivo studies with Tiopronin 13C D3 focus on pharmacokinetic characterization. Rats or mice are administered tiopronin, and plasma concentrations are measured by LC-MS/MS using the stable isotope-labeled internal standard. Parameters such as Cmax, Tmax, AUC, t1/2, and clearance are determined.
These protocols are for reference only. InvivoChem does not independently validate these methods.

(Please use freshly prepared in vivo formulations for optimal results.)
Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 5.9801 mL 29.9007 mL 59.8015 mL
5 mM 1.1960 mL 5.9801 mL 11.9603 mL
10 mM 0.5980 mL 2.9901 mL 5.9801 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly.
