TCO-PEG4-NHS ester is a PEG-based PROTAC linker and a cleavable ADC linker used in the synthesis of antibody-drug conjugates (ADCs). It contains a trans-cyclooctene (TCO) group, a PEG4 spacer, and an N-hydroxysuccinimide (NHS) ester. TCO-PEG4-NHS ester has a molecular formula of C24H38N2O10, a molecular weight of 514.57 g/mol, and a CAS number of 1613439-69-2.
Biological Activity I Assay Protocols (From Reference)
Targets
As a chemical linker, TCO-PEG4-NHS ester does not have a specific biological target. It is used for the modification of biomolecules through NHS ester-mediated conjugation to primary amines. The PEG4 spacer improves water solubility, and the TCO group enables click chemistry reactions with tetrazines. The compound is used in PROTAC and ADC synthesis.
ln Vitro
Two distinct ligands, one for the E3 ubiquitin ligase and the other for the target protein, are present in PROTAC and are joined by a linker. PROTAC targets and selectively degrades target proteins by means of the intracellular ubiquitin-proteasome system. An ADC cytotoxin is connected to an antibody by use of an ADC linker to form an ADC.
In vitro activity is not assessed for this compound as it is a chemical linker rather than a bioactive molecule. Its chemical reactivity and utility in bioconjugation, PROTAC synthesis, and ADC synthesis are the primary focus.
ln Vivo
In vivo activity is not applicable for TCO-PEG4-NHS ester as it is not a pharmacologically active compound. The compound is used in research settings as a chemical linker for bioconjugation and drug delivery applications. It is not intended for therapeutic use.
Enzyme Assay
In vitro enzyme/receptor binding assays are not typically performed for TCO-PEG4-NHS ester, as it is a chemical linker rather than a bioactive compound. Its reactivity and conjugation efficiency are characterized by standard analytical techniques.
Cell Assay
In vitro cell-based assays are not applicable for TCO-PEG4-NHS ester as it is not a pharmacologically active compound. The compound is used in chemical synthesis and bioconjugation and is not evaluated for its effects on cell viability or other cellular functions.
Animal Protocol
In vivo animal studies are not performed for TCO-PEG4-NHS ester as it is not a pharmacologically active compound. The compound is used as a chemical linker and is not intended for therapeutic use. Any animal studies involving this compound would be limited to evaluating the pharmacokinetics of TCO-PEG4-conjugated molecules.
ADME/Pharmacokinetics
Pharmacokinetic properties of TCO-PEG4-NHS ester are not characterized as the compound is not a drug candidate. The compound is water-soluble and can be easily conjugated to biomolecules such as proteins, peptides, and antibodies for imaging, targeting, and drug delivery applications. It is stored at appropriate temperatures for research use.

Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 1.9434 mL 9.7169 mL 19.4337 mL
5 mM 0.3887 mL 1.9434 mL 3.8867 mL
10 mM 0.1943 mL 0.9717 mL 1.9434 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
