Biological Activity I Assay Protocols (From Reference)
Targets
TC HSD 21 is a potent 17β-hydroxysteroid dehydrogenase type 3 (17β-HSD3) inhibitor with an IC50 of 14 nM. IC50 values are 6 and 40 nM at human and mouse 17β-HSD3, respectively. It displays no activity at 17β-HSD1. 17β-HSD3 is a crucial enzyme in the final step of testosterone synthesis in the testes, catalyzing the conversion of androstenedione to testosterone. The compound also shows excellent selectivity over 17β-HSD isoenzymes and nuclear receptors.
ln Vitro
In vitro, TC HSD 21 demonstrates potent inhibition of 17β-HSD3 enzyme activity. It is a cell-permeable compound, making it suitable for cellular assays. It shows excellent selectivity over other 17β-HSD isoenzymes and nuclear receptors. The compound is used to dissect androgen biosynthesis and 17β-HSD3-dependent pathways in cell-based systems.
ln Vivo
In vivo, TC HSD 21 is used to study the role of testicular androgen production in disease models. By inhibiting 17β-HSD3, it blocks testosterone synthesis in the testes. This makes it a valuable tool for studying androgen-dependent diseases and endocrine regulation. It regulates glucocorticoid metabolism by blocking local cortisol regeneration, thereby affecting glucose metabolism and inflammatory responses.
Enzyme Assay
For non-cellular enzyme/receptor binding assays, the compound's inhibition of 17β-HSD3 can be assessed using enzyme activity assays with purified 17β-HSD3 protein. The conversion of androstenedione to testosterone is measured in the presence of varying concentrations of TC HSD 21. IC50 values are determined. Selectivity over other 17β-HSD isoenzymes and nuclear receptors can be assessed using similar enzyme activity assays or radioligand binding assays.
Cell Assay
For in vitro cell-based assays, cells expressing 17β-HSD3 are cultured and treated with TC HSD 21. Testosterone production is measured to assess enzyme inhibition. Cell permeability allows for intracellular target engagement. The compound can be used to study the effects of 17β-HSD3 inhibition on androgen biosynthesis and downstream signaling pathways in various cell types.
Animal Protocol
For in vivo animal studies, TC HSD 21 is administered to animal models to study steroid metabolism and endocrine regulation. It can be used to investigate the role of testicular androgen production in disease models. By blocking local cortisol regeneration, it affects glucose metabolism and inflammatory responses. Dosing regimens and routes of administration depend on the specific experimental model.
ADME/Pharmacokinetics
Pharmacokinetic properties include solubility in DMSO (10 mM * 1 mL in DMSO ready for reconstitution). The powder is stable at -20°C for 3 years and at 4°C for 2 years. In solvent, it is stable at -80°C for 6 months and at -20°C for 1 month. The appearance is a solid, light yellow to yellow. The molecular weight is 422.32.
Toxicity/Toxicokinetics
In preclinical studies, TC HSD 21 has shown good selectivity and potency. No significant toxicity has been reported. It is a controlled substance and not for sale in some territories. The compound is for research use only and not for human use.
