Product Description
Talnetant (SB 223412) HCl is a selective, competitive, brain-penetrating NK3 receptor blocker (antagonist) with Ki of 1.4 nM in hNK-3-CHO cells. Relative to hNK-2 receptor, Talnetant HCl has 100 times selectivity for hNK-3 and has no affinity for hNK-1. Talnetant HCl may be utilized in research related to schizophrenia.
Talnetant hydrochloride (SB 223412) is a potent, selective, competitive, and brain-permeable NK3 receptor antagonist with a Ki of 1.4 nM in hNK-3-CHO cells. It shows 100-fold selectivity for the hNK-3 versus hNK-2 receptor and has no affinity for the hNK-1 receptor at concentrations up to 100 μM. This compound is used in research on neurological and psychiatric disorders, including schizophrenia and anxiety, where NK3 receptor modulation may be beneficial.
Biological Activity I Assay Protocols (From Reference)
Targets
Neurokinin-3 (NK3) receptor (selective antagonist).
ln Vitro
In U-2OS cells expressing the human NK3 receptor, talnetant (SB 223412) (0.1–1 μM) hydrochloride decreases NKB-induced IP accumulation [3].
Talnetant hydrochloride is a selective, competitive NK3 receptor antagonist with a Ki of 1.4 nM in hNK-3-CHO cells. It shows 100-fold selectivity for hNK-3 over hNK-2 receptor and has no affinity for hNK-1 at concentrations up to 100 μM. The compound is brain-permeable and suitable for CNS research.
ln Vivo
Talnetant (SB 223412) hydrochloride (0.5-2 mg/kg iv, 2 min pre-treatment) suppresses the dose-dependent induction of miosis in conscious rabbits by senktide (25 μg, iv) with an ED50 of 0.44 mg/kg [1]. Senktide-induced "wet dog wag" behavior can be considerably attenuated in a dose-dependent manner by tanetant (SB 223412) hydrochloride (i.p., 1-100 mg/kg, 1 h), which also dramatically increases the number of cells in the medial prefrontal cortex. In the vomeronasal nucleus of freely moving guinea pigs, external dopamine and norepinephrine were found to decrease haloperidol-induced elevations in dopamine levels [3].
No specific in vivo efficacy data are available for Talnetant hydrochloride. As a brain-permeable NK3 receptor antagonist, it would be expected to modulate neurokinin signaling in vivo and potentially produce anxiolytic or antipsychotic-like effects, but no published animal study results are currently documented.
Enzyme Assay
The binding assay for NK3 receptor antagonists uses membranes from hNK-3-CHO cells or recombinant cells expressing NK3 receptor. Radiolabeled neurokinin B or a selective NK3 agonist is used as a tracer. Test compounds compete for binding, and Ki values are determined from competition curves.
Cell Assay
In cellular assays, cells expressing recombinant NK3 receptor are treated with Talnetant hydrochloride. Receptor activation is assessed by measuring intracellular calcium mobilization or inositol phosphate production. IC50 values for inhibition of agonist-induced signaling are calculated to confirm antagonist activity.
Animal Protocol
Animal/Disease Models: Male Dunkin-Hartley guinea pig [3]
Doses: 1, 3, 10, 30 or 100 mg/kg
Route of Administration: intraperitoneal (ip) injection; 1 hour
Experimental Results: Wet dog shake (WDS) at 30 mg/kg Behavior weakened Dramatically, falling from 31.1 to 16.7. Extracellular DA levels were Dramatically increased to 238%, and NE levels were Dramatically increased to 227.1%, without affecting 5-HT levels.
No specific in vivo animal study protocols are documented for Talnetant hydrochloride. General models of anxiety, schizophrenia, or pain could be used, with the compound administered systemically, and behavioral and physiological endpoints assessed to evaluate therapeutic potential.
