Product Description
Suloctidil HCl is a novel and potent Vasodilator gent
Suloctidil HCl (CAS 54767-71-4) is the hydrochloride salt of suloctidil, a peripheral vasodilator formerly used in the management of peripheral and cerebral vascular disorders. With molecular formula C20H36ClNOS and molecular weight 374.02 g/mol, it acts as a Na⁺/K⁺ ATPase inhibitor and modulates membrane fluidity. It is hepatotoxic and fatalities have occurred, limiting its clinical use. It is used as a research tool for studying vascular pharmacology and Na⁺/K⁺ ATPase inhibition.
Biological Activity I Assay Protocols (From Reference)
Targets
The primary target of Suloctidil HCl is the Na⁺/K⁺ ATPase, a membrane-bound ion pump responsible for maintaining electrochemical gradients. It acts as an inhibitor of Na⁺/K⁺ ATPase activity and modulates membrane fluidity. By inhibiting this pump, it affects vascular smooth muscle tone and peripheral blood flow. It is a peripheral vasodilator that was formerly used for vascular disorders.
ln Vitro
In vitro, Suloctidil HCl is a peripheral vascular dilator and Na⁺/K⁺ ATPase inhibitor. It modulates membrane fluidity in various cell types. Specific IC50 values for Na⁺/K⁺ ATPase inhibition are reported in the literature. It is used as a reference compound for studying vascular pharmacology. It is typically tested at concentrations ranging from 1 to 100 µM.
ln Vivo
In vivo, Suloctidil HCl was formerly used in the management of peripheral and cerebral vascular disorders. It is hepatotoxic and fatalities have occurred, leading to its withdrawal. Specific published in vivo efficacy data, including dosing regimens and clinical outcomes, are reported in clinical literature. It is primarily used as a research tool for studying Na⁺/K⁺ ATPase inhibition and vascular pharmacology.
Enzyme Assay
The Na⁺/K⁺ ATPase inhibition assay uses purified enzyme from kidney or brain tissue or recombinant enzyme expressed in cells. The enzyme is incubated with ATP and varying concentrations of Suloctidil HCl in reaction buffer. ATP hydrolysis is measured by quantifying inorganic phosphate release using a colorimetric assay. IC50 values are calculated from dose-response curves.
Cell Assay
For cell-based assays, vascular smooth muscle cells or endothelial cells are treated with Suloctidil HCl. Na⁺/K⁺ ATPase activity is measured in cell lysates using ATP as substrate. Membrane fluidity is assessed using fluorescence polarization with DPH or TMA-DPH probes. Cell viability is evaluated using MTT or LDH release assays.
Animal Protocol
In vivo studies with Suloctidil HCl employed rodent models for vascular assessment. The compound is dissolved in saline and administered intraperitoneally or orally. Key readouts include blood pressure, peripheral blood flow, and Na⁺/K⁺ ATPase activity in tissues. Due to its hepatotoxicity, in vivo studies are conducted with appropriate safety precautions.

Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 2.6737 mL 13.3683 mL 26.7365 mL
5 mM 0.5347 mL 2.6737 mL 5.3473 mL
10 mM 0.2674 mL 1.3368 mL 2.6737 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly.
