Product Description
Sonepiprazole mesylate is a novel and potent D4 antagonist
Sonepiprazole mesylate (CAS 170858-34-1), also known as U-101387G or PNU-101387G, is a selective dopamine D4 receptor antagonist. With a molecular formula of C22H31N3O6S2 and molecular weight of 497.63 g/mol, Sonepiprazole mesylate exhibits inhibition constants (Kis) of 3.6 nM for rD4-Dopamine and 10.1 nM for hD4.2-Dopamine, indicating high selectivity for the D4 receptor subtype. It shows much lower affinity for D2, D3, and other dopamine receptor subtypes. Sonepiprazole mesylate is investigated for its potential in treating schizophrenia and other psychiatric disorders.
Biological Activity I Assay Protocols (From Reference)
Targets
The primary target of Sonepiprazole mesylate is the dopamine D4 receptor, a G protein-coupled receptor that is highly expressed in the prefrontal cortex, hippocampus, and limbic system. It acts as a selective antagonist with Ki values of 3.6 nM for rat D4 and 10.1 nM for human D4.2 receptors. The compound shows high selectivity for D4 over D2 (Ki = 5147 nM) and D3 (Ki = 7430 nM) receptors. By blocking D4 receptors, Sonepiprazole mesylate modulates dopaminergic signaling in brain regions involved in cognition, emotion, and reward.
ln Vitro
In vitro, Sonepiprazole mesylate is a selective D4 dopamine receptor antagonist with Ki values of 3.6 nM for rD4-Dopamine and 10.1 nM for hD4.2-Dopamine. It shows >1,000-fold selectivity for D4 over D2 and D3 receptors. The compound is used in radioligand binding assays to characterize D4 receptor pharmacology and in functional assays of dopamine-mediated signaling. It is a valuable tool for studying the role of D4 receptors in psychiatric disorders and cognitive function.
ln Vivo
In vivo, Sonepiprazole mesylate has been evaluated in animal models of schizophrenia and other psychiatric disorders. As a selective D4 antagonist, it may modulate dopaminergic neurotransmission in brain regions implicated in psychosis. Preclinical studies assess its effects on behaviors including locomotor activity, prepulse inhibition, and cognitive performance. Specific published in vivo efficacy data, including dosing regimens and behavioral outcomes, are reported in preclinical literature.
Enzyme Assay
The dopamine D4 receptor binding assay uses membranes from cells expressing recombinant D4 receptors. Radiolabeled dopamine or a selective D4 antagonist (e.g., [3H]spiperone) is incubated with membranes and Sonepiprazole mesylate at serial dilutions. Bound radioactivity is separated by filtration and quantified by scintillation counting. Ki values are calculated from competition curves. Selectivity is assessed against D2, D3, and other dopamine receptor subtypes.
Cell Assay
For cell-based assays, cells expressing D4 receptors are treated with Sonepiprazole mesylate. Receptor activation is assessed by measuring inhibition of forskolin-stimulated cAMP accumulation or by G-protein-coupled inwardly rectifying potassium (GIRK) channel currents using patch-clamp electrophysiology. Dopamine-induced calcium mobilization is measured using fluorescent indicators. Cell viability is evaluated using standard reagents.
Animal Protocol
In vivo studies with Sonepiprazole mesylate employ rodent models of psychiatric disorders. The compound is dissolved in saline or vehicle and administered intraperitoneally or orally. Doses are selected based on pharmacokinetic studies. Key readouts include locomotor activity, prepulse inhibition of startle response, and cognitive performance in maze tasks. Brain tissue is collected for receptor occupancy studies and neurochemical analysis.
