Product Description
Razobazam is a novel and potent drug of benzodiazepine analogue
Razobazam (CAS: 78466-98-5) is a benzodiazepine derivative with cognitive-enhancing (nootropic) activity. Also known as Hoe 175, it has been shown to improve learning performance in socially deprived rats. Molecular formula is C₁₄H₁₄N₄O₂ and molecular weight is 270.29.
Biological Activity I Assay Protocols (From Reference)
Targets
Razobazam targets the central nervous system to enhance cognitive function. As a benzodiazepine derivative, it likely interacts with GABA-A receptors or related CNS targets, though its specific binding site is not fully characterized. It produces nootropic effects in animal studies.
ln Vitro
In vitro, razobazam demonstrates effects on neuronal activity consistent with cognitive enhancement. It has been studied in brain tissue for its effects on neuronal density and metabolic activity. The compound increases neuronal density in the frontal cortex by 13%.
ln Vivo
In vivo, razobazam improves learning performance in socially deprived rats, showing an 18% increase in avoidance scores post-training. It induces significant changes in brain optical density, including a 22% decrease in the lateral habenula, a 25% increase in the ventral tegmental area, and a 13% increase in the prefrontal cortex.
Enzyme Assay
In vitro receptor binding assays use membrane preparations from brain tissue expressing GABA-A receptors or other benzodiazepine targets. Radiolabeled ligands such as [³H]flunitrazepam are used as tracers. Varying concentrations of razobazam are incubated with membranes, and bound radioactivity is measured by filtration or scintillation counting.
Cell Assay
In vitro cell-based assays evaluate razobazam in neuronal cell cultures. Cells are treated with the compound, and neuronal activity, calcium signaling, or receptor modulation is assessed. Cytotoxicity is evaluated using standard viability assays. Receptor binding studies may use receptor-expressing cell lines.
Animal Protocol
In vivo animal studies use socially deprived rat models. Rats are administered razobazam, and learning performance is assessed using active avoidance tests. Brain metabolic activity is measured by 2-deoxyglucose uptake. Optical density changes in specific brain regions (lateral habenula, ventral tegmental area, prefrontal cortex) are quantified.

Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 3.6997 mL 18.4986 mL 36.9973 mL
5 mM 0.7399 mL 3.6997 mL 7.3995 mL
10 mM 0.3700 mL 1.8499 mL 3.6997 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly.
