| Targets |
The primary target is intracellular calcium ions (Ca²⁺). Quin-2 is a fluorescent chelator that binds calcium with high affinity (Kd 115 nM). The AM ester form is cell-permeant and is hydrolyzed by intracellular esterases to trap the active dye inside cells. Upon calcium binding, Quin-2 exhibits an increase in fluorescence, allowing measurement of intracellular calcium concentrations.

1. Working solution preparation for Quin-2AM 1.1 Stock solution preparation: Extract Quin-2AM with anhydrous DMSO to create a 10 mM stock solution. Note: It is advised to aliquot the Quin-2AM storage solution and keep it at -20°C or -80°C in the dark. 1.2 Configuring the working solution: To create a 1-10 μM Quin-2AM working solution, add the current solution to a previously prepared HBSS. Note: Please use the present preparation and modify the Quin-2AM working solution concentration based on the actual circumstances. 2. Suspended cell staining (2.1): Centrifuge cells, add PBS, then wash twice for five minutes each time. Add 1 milliliter of Quin-2AM working solution and refrigerate for five to thirty minutes until the cell density reaches 1×106/mL 2.2. 2.3 Centrifuge for 3–4 minutes at 400 g, then remove the supernatant. 2.4 Wash the cells twice with PBS, giving them five minutes each time. 2.5 Re-suspend the cells in 1 milliliter of HBSS and use a flow cytometer or fluorescence microscope to observe. 3. Adherent cell staining (3.1): Grow adherent cells on sterile coverslips. 3.2 Take off the coverslip from the cells and use a little aspirator to extract a few cells. 3.3 Finish filling by adding 100 μL of dye working solution and shaking. 3.4 Aspirate the dye working solution, give it two or three 5-minute culture medium washes, and examine under a fluorescence microscope.
Quin-2AM is a fluorescent Ca²⁺ chelator with high affinity for calcium (Kd 115 nM). It displays high selectivity for calcium over other divalent cations. It is cell-permeant and shows low cytotoxicity. It has been used to measure intracellular calcium in Caco-2 cells and other cell types. It shows less compartmentalization than Fura-2 in some cell types. |
| ln Vivo |
In vivo data are not applicable, as Quin-2AM is a fluorescent indicator used for in vitro calcium imaging. It is not administered to animals. It is used in isolated cells and tissues for calcium measurement.
|
| Enzyme Assay |
Quin-2AM is loaded into cells by incubation at 37°C for 15-60 min in culture medium. The AM ester is hydrolyzed by intracellular esterases to release Quin-2. Cells are washed to remove extracellular dye. Fluorescence is measured using a fluorescence plate reader or spectrofluorometer at excitation 339 nm and emission 492 nm. Calcium concentrations are calculated from fluorescence intensities.
|
| Cell Assay |
Cell-based calcium imaging assays use cells grown on coverslips or in 96-well plates. Cells are loaded with Quin-2AM (1-20 µM) in culture medium containing pluronic acid for 30-60 min at 37°C. After washing, cells are stimulated with agonists, and fluorescence is monitored using a fluorescence microscope or plate reader. Calcium responses are quantified.
|
| Animal Protocol |
Not applicable. Quin-2AM is not used in animal experiments. Its application is limited to in vitro calcium imaging in isolated cells and tissues.

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). *Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
