Hydroxy-PEG4-methyl acetate is a PEG-based PROTAC linker consisting of a hydroxyl group (OH) and a methyl acetate (carboxylic acid methyl ester) group connected by a 4-unit PEG spacer (PEG4). ln Vitro
One ligand is for an E3 ubiquitin ligase, and the other is for the target protein; these two ligands are joined by a linker to form PROTACs. The intracellular ubiquitin-proteasome system is utilized by PROTACs to specifically destroy target proteins[1].
As a linker molecule, Hydroxy-PEG4-methyl acetate itself has no intrinsic biological activity; it serves as a structural connector to join a target protein ligand and an E3 ubiquitin ligase ligand in PROTAC synthesis. The methyl ester group can be hydrolyzed under basic conditions (e.g., with LiOH) to a carboxylic acid, which can then be activated with EDC/NHS to form an amide bond with an amine-containing ligand. The hydroxyl group can be used for further functionalization, such as tosylation or conversion to an amine, or can be protected (e.g., as a benzyl ether) for orthogonal deprotection strategies. The combination of a hydroxyl group and a methyl ester provides orthogonal functional groups for bioconjugation.
