| Targets |
Hydroxylapatite does not have a defined pharmacological target. Its mechanism of action is physical and biological: it provides a scaffold for bone cell attachment, proliferation, and differentiation. The compound binds to bone tissue and promotes osseointegration. It is also used as a chromatographic medium for protein purification.
|
|---|---|
| ln Vitro |
In vitro, hydroxylapatite is used as a biomaterial for cell culture studies. It serves as a substrate for osteoblast adhesion, proliferation, and differentiation. The compound is also used as a chromatographic medium for the separation of proteins, nucleic acids, and other biomolecules. Its biocompatibility and osteoconductivity are assessed using standard cell-based assays.
|
| ln Vivo |
In vivo, hydroxylapatite is used as a bone graft substitute and as a coating for dental and orthopedic implants. It promotes bone regeneration and osseointegration by providing a scaffold for bone cell attachment and growth. The compound is biocompatible and integrates well with surrounding bone tissue.
|
| Enzyme Assay |
For in vitro cell-based experiments, hydroxylapatite is used as a scaffold for osteoblast culture. Osteoblast-like cells (e.g., MC3T3-E1) are seeded onto hydroxylapatite discs or particles and cultured for 7-21 days. Cell adhesion is assessed by scanning electron microscopy (SEM). Cell proliferation is measured using MTT or DNA quantification assays. Osteogenic differentiation is evaluated by measuring alkaline phosphatase (ALP) activity and mineralization (alizarin red staining).
|
| Cell Assay |
For in vitro cell-based experiments, hydroxylapatite can also be used to assess its effects on immune cells. Macrophages are cultured with hydroxylapatite particles, and cytokine production (e.g., TNF-α, IL-6, IL-10) is measured by ELISA to evaluate the inflammatory response. The compound's biocompatibility is assessed by measuring cell viability using standard assays.
|
| Animal Protocol |
In vivo animal studies using hydroxylapatite are conducted in models of bone defect repair. Hydroxylapatite scaffolds or granules are implanted into bone defects in rodents, rabbits, or larger animals. Bone regeneration is assessed by micro-CT, histology, and histomorphometry at various time points (e.g., 4-12 weeks). Osseointegration of hydroxylapatite-coated implants is evaluated by biomechanical testing and histological analysis.
|
| ADME/Pharmacokinetics |
Pharmacokinetic properties of hydroxylapatite are not applicable, as it is a biomaterial that is not administered systemically. When implanted, hydroxylapatite undergoes gradual resorption and remodeling by osteoclasts, releasing calcium and phosphate ions that are incorporated into new bone tissue. The compound is not absorbed into the systemic circulation.
|
| Toxicity/Toxicokinetics |
Hydroxylapatite is biocompatible and non-toxic. It is widely used in medical devices and bone graft materials. The compound is not classified as hazardous. Standard laboratory safety practices should be followed when handling the compound. Hydroxylapatite is for research and medical use, not for human therapeutic applications as a drug.

| Additional Infomation |
Hydroxyapatite is a mineral with the chemical formula Ca5(PO4)3OH. Its corresponding International Mineralogical Association (IMA) number is IMA2010 sp, and its IMA symbol is Hap. It is a mineral component of bones and teeth; in treatment, it has been used as a prosthetic material and for the prevention and treatment of osteoporosis. Therapeutic Uses Experimental Uses: Female patients with primary cholestasis who exhibit poor absorption of calcium, phosphate, and vitamin D, and experience accelerated thinning of cortical bone. The value of hydroxyapatite in treating cortical bone thinning in primary cholestasis was evaluated. Cortical bone loss was observed in the control group. Cortical bone thickness was significantly increased in the hydroxyapatite group. Hydroxylapatite (Hydroxyapatite, CAS 1306-06-5) is a biomaterial and medical device component, not an FDA-approved pharmaceutical drug. Its primary applications are in bone graft substitutes, dental implants, orthopedic coatings, and as a chromatographic medium for protein purification. No clinical trials or approved therapeutic indications exist for this compound as a drug. |

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations
Oral Formulation 1: Suspend in 0.5% CMC Na (carboxymethylcellulose sodium) Oral Formulation 2: Suspend in 0.5% Carboxymethyl cellulose Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals).
