ln Vitro
In vitro, Peptone from casein promotes microbial growth and proliferation. It is tested for its ability to support the growth of various indicator organisms (e.g., Escherichia coli, Staphylococcus aureus, Pseudomonas aeruginosa, Candida albicans) in solid or liquid culture media. Cell Assay
Microbial growth promotion test protocol: Test microorganisms (e.g., E. coli ATCC 8739, S. aureus ATCC 6538, P. aeruginosa ATCC 9027, C. albicans ATCC 10231) are streaked onto agar plates containing casein peptone as the nitrogen source, or inoculated into broth media. Plates are incubated at 30-37degC for 18-48 hours, and colony morphology, growth density, and colony size are compared to reference media. For quantitative assessment, optical density at 600 nm is measured after 18-24 hours of growth in liquid culture.
Animal Protocol
Animal studies using casein peptone are not typical for drug development purposes. In specialized research (e.g., acute inflammation models), sterile casein peptone solution (e.g., 10% w/v) may be injected intraperitoneally into rodents (0.5-1 mL per mouse) to induce neutrophil recruitment. Animals are sacrificed at various time points (e.g., 4-24 hours), and peritoneal exudate is collected by lavage for differential cell counting and cytokine analysis. This model is used to study inflammatory cell migration and sterile peritonitis.
