CI-1040 hydrochloride can directly inhibit MEK1 with an IC50 value of 17 nM. Studies have shown that it has low activity against a series of related kinases, with an IC50 value more than 2.5 orders of magnitude higher than that of MEK1. Treatment of intact cells with CI-1040 hydrochloride can completely inhibit mitogen-stimulated ERK phosphorylation. In MDA-MB-231 breast cancer cells, a concentration of 1 μM CI-1040 hydrochloride can inhibit ERK1 and ERK2 phosphorylation by 99% and 92%, respectively [1]. CI-1040 hydrochloride induces apoptosis and inhibits proliferation of U-937 cells in a dose- and time-dependent manner. CI-1040 hydrochloride can significantly increase PUMA mRNA and protein levels [2].

Systemic application of the MEK inhibitor CI-1040 hydrochloride reduced adenoma formation by one-third and significantly restored lung structure. Lung cell proliferation was reduced in mice treated with CI-1040 hydrochloride, but it had no significant effect on alveolar cell differentiation[3].
