Canagliflozin hydrochloride effectively inhibits human dipeptidyl peptidase IV (DPP-IV) with an IC50 value of 6.8 nM [1]. 10 μM canagliflozin hydrochloride is highly selective for human DPP-IV, with no significant off-target activity at a concentration of 10 μM, and its selectivity for related proline-specific dipeptidyl peptidases is more than 100 times higher [1]. In vitro experiments show that canagliflozin hydrochloride does not inhibit or induce CYP450 enzymes [1].

Canagliflozin (0.3 mg/kg; orally; single dose) hydrochloride reduced blood glucose fluctuations by 66% 40 minutes after oral glucose tolerance test in insulin-resistant Zucker's obese (fa/fa) rats[1]. Canagliflozin (20 mg/kg; orally; once daily; for 7 days) hydrochloride improved insulin sensitivity by increasing glucose infusion rate (GIR) and showed a trend toward reducing hepatic glycogen production in insulin-resistant Zucker's obese (fa/fa) rats[1]. Canagliflozin (10 mg/kg; orally; single dose) hydrochloride significantly reduced fasting blood glucose and decreased AUC0-t by 30% in oral glucose tolerance test[1]. Canagliflozin (3 mg/kg; orally; single dose) hydrochloride produced sustained plasma DPP-IV inhibition in non-diabetic cynomolgus monkeys, retaining 40% and 60% of baseline activity at 24 and 48 hours after administration, respectively[1].
