| Targets |
Bis(2-chloroethyl) phosphate has no therapeutic target. It is a metabolite and an environmental pollutant. It is a suspected carcinogen. It is formed in the body by the metabolism of TCEP. The parent compound, TCEP, is a neurotoxicant and has been shown to cause hippocampal brain lesions. The mechanism of toxicity may involve the disruption of lipid metabolism and cholinergic signaling. BCEP is not an enzyme inhibitor for therapeutic purposes.
|
|---|---|
| ln Vitro |
Bis(2-chloroethyl) phosphate is not a drug and has no direct in vitro biological activity as a therapeutic agent. It is a urinary metabolite of TCEP. The cytotoxicity (LC₅0) of BCEP was measured as 456 microM in primary mouse hepatocytes (PMH). This indicates it is moderately toxic. It is used as an analytical standard for the quantification of organophosphate flame retardants (OPFRs) in urine. It can also be used in in vitro metabolism studies to measure the activity of enzymes that hydrolyze TCEP. No specific IC₅0 values for receptor binding are available.
|
| ln Vivo |
In vivo, Bis(2-chloroethyl) phosphate is not used as a drug. It is a metabolite detected in the urine of humans and animals after exposure to TCEP. The median concentration in human urine is 0.68 ng/mL. It is a biomarker for assessing exposure to TCEP in occupational settings and the general population. Elevated levels of BCEP are associated with potential health risks, including hepatotoxicity and neurotoxicity. It is not a therapeutic agent.

| Enzyme Assay |
In vitro toxicity assay: Primary mouse hepatocytes (PMHs) are seeded in 96-well plates (1×10⁴ cells/well). After 24 h, the cells are treated with various concentrations of Bis(2-chloroethyl) phosphate (0-1000 uM) for 24 h. Cell viability is measured by the MTT assay or CCK-8 assay. The half-maximal lethal concentration (LC₅0) is calculated (456 uM). To measure metabolism, microsomal incubations with TCEP are performed, and BCEP formation is quantified by LC-MS/MS. For environmental analysis, water samples (1 L) are extracted with solid-phase extraction (SPE) (Oasis HLB cartridges) and analyzed by LC-MS/MS.

| Cell Assay |
Bis(2-chloroethyl) phosphate is not used in standard cell-based assays for drug discovery. For toxicity screening, HepG2 or HEK293 cells are seeded in 96-well plates (1×10⁴ cells/well) and treated with Bis(2-chloroethyl) phosphate (1-1000 uM) for 24-72 h. Cell viability is measured by MTT assay. The CC₅0 is expected to be > 200 uM, indicating moderate cytotoxicity. For metabolism studies, primary human hepatocytes are seeded in 6-well plates, treated with TCEP (10-100 uM), and the appearance of BCEP in the medium is measured by LC-MS/MS. The intrinsic clearance (Clint) is calculated.
|
| Animal Protocol |
For toxicokinetic studies, male Sprague-Dawley rats (200-250 g, n=6/group) are administered TCEP orally (10-100 mg/kg) in corn oil. Blood samples (0.5 mL) are collected by tail vein at 0, 0.5, 1, 2, 4, 6, 12, 24 h post-dose. Urine is collected in metabolic cages for 24 h. The concentration of Bis(2-chloroethyl) phosphate in plasma and urine is measured by LC-MS/MS. The half-life (t½) of TCEP in plasma is 2-4 h. The fraction of the dose excreted as BCEP is calculated (< 5%). The compound is not administered directly.

| ADME/Pharmacokinetics |
Bis(2-chloroethyl) phosphate (MW 222.99, Log P 0.5) is a polar, negatively charged molecule. It has low oral bioavailability due to its high water solubility. It is not metabolized further. It is rapidly excreted in the urine. The half-life of elimination in rats is 2-4 h. The compound is a major metabolite of TCEP. It is not intended for therapeutic use. For research use, it is stored as a solid at -20degC. It is soluble in water and DMSO.

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals).
