Stable heavy isotopes of hydrogen, carbon, and other elements have been incorporated into drug molecules, largely as tracers for quantitation during the drug development process. Studies involving the human use of drugs labeled with deuterium suggest that these compounds may offer some advantages when compared with their nondeuterated counterparts. Deuteration has gained attention because of its potential to affect the pharmacokinetic and metabolic profiles of drugs. Deutetrabenazine is the first deuterated drug to receive Food and Drug Administration approval. This deuterated form of the drug tetrabenazine is indicated for the treatment of chorea associated with Huntington's disease as well as tardive dyskinesia. Ongoing clinical trials suggest that a number of other deuterated compounds are being evaluated for the treatment of human diseases and not merely as research tools.[1] Treatment of LPS-stimulated microglia with 5, 10, and 20 μM Ribavirin (ICN-1229) induced a 43% (p<0.05), 53% (p<0.05), and 59% (p < 0.05) decrease in NO2 levels. Ribavirin (ICN-1229) (10 mM) did not significantly reduce cell surface area in non-stimulated cultures, but significantly reduced cell surface area in LPS-stimulated microglia (32%, p<0.05)[4]. Ribavirin (ICN-1229) was active against DENV with an EC50 of 3 μM in A549 cells, and the combination of CM-10-18 and Ribavirin (ICN-1229) was effective in reducing viral replication[5].

ALT, AST activities and bilirubin levels were significantly decreased by JAT combined with interferon and Ribavirin (ICN-1229) administration (p<0.01). When JAT, interferon, or Ribavirin were used alone with CCl4, the liver appeared to exhibit some hepatoprotection against CCl4 as evidenced by the presence of normal hepatic cords, the absence of necrosis, and less fat infiltration. Groups treated with JAT, pegylated interferon, and Ribavirin (ICN-1229) alone or in combination showed decreased expression of TGF-β and Bax. In the interferon, Ribavirin (ICN-1229) and JAT triple treatment group, the expression level of p53 was significantly reduced[2]. Wistar rats treated with Ribavirin (ICN-1229) capsules (400 mg Ribavirin) showed a significant decrease in activin A and a significant increase in follistatin at serum and liver levels. Ribavirin (ICN-1229) exhibits strong antiviral activity only when it is used in combination with IFN-α or Peg-IFN-α[3]. Ribavirin (40 mg/kg, po) significantly enhanced the antiviral efficacy of CM-10-18 in mice. Ribavirin (ICN-1229) inhibits DENV infection in cultured cells, but single-agent treatment is ineffective in reducing viremia[5].
