Cells are seeded in 24-well plates at 5 × 10⁴ cells per well and grown to 70-80% confluence. Cells are serum-starved for 24 hours and then treated with Palmitoyl Tripeptide-8 at concentrations of 1, 10, and 100 μg/mL for 24-72 hours. Culture supernatants are collected for collagen quantification by ELISA (human type I collagen, type III collagen). Cells are lysed for RNA extraction and qRT-PCR analysis of collagen genes (COL1A1, COL3A1) and other extracellular matrix genes (FN1, ELN, MMP1). For cell proliferation assays, cells are seeded in 96-well plates and treated with the peptide for 24-72 hours, and cell viability is assessed using the MTT or CellTiter-Glo assay.
