Enzyme Assay
For in vitro assays with Palmitoyl Tripeptide-8, the following protocols are used: For collagen synthesis assays, human dermal fibroblasts (HDFs) are cultured in DMEM with 10% FBS at 37°C in 5% CO₂. Cells are seeded in 24-well plates at 5 × 10⁴ cells per well and grown to 70-80% confluence. Cells are serum-starved for 24 hours and then treated with Palmitoyl Tripeptide-8 at concentrations of 1, 10, and 100 μg/mL for 24-72 hours. Culture supernatants are collected for collagen quantification by ELISA (human type I collagen, type III collagen). Cells are lysed for RNA extraction and qRT-PCR analysis of collagen genes (COL1A1, COL3A1) and other extracellular matrix genes (FN1, ELN, MMP1). For cell proliferation assays, cells are seeded in 96-well plates and treated with the peptide for 24-72 hours, and cell viability is assessed using the MTT or CellTiter-Glo assay. For anti-allergen assays, human mast cells (e.g., LAD2 cells) or RBL-2H3 cells are sensitized with IgE and then stimulated with antigen (e.g., DNP-BSA) in the presence or absence of the peptide. Histamine release is measured by ELISA. Cytokine release (TNF-α, IL-6, IL-8) is measured by ELISA. For UV protection studies, fibroblasts or keratinocytes are treated with the peptide and then exposed to UV radiation (UVA or UVB), and cell viability and ROS production are assessed.
Animal Protocol
For in vivo clinical studies with Palmitoyl Tripeptide-8, the following general protocol is used: human subjects (30-60 years old) with mild to moderate signs of skin aging are enrolled in a randomized, double-blind, vehicle-controlled study. The test product containing 0.1-1% Palmitoyl Tripeptide-8 and the vehicle control are applied topically to designated areas (e.g., face, forearm) twice daily for 4-12 weeks. Skin parameters are assessed at baseline and at regular intervals (2, 4, 8, 12 weeks). Skin hydration is measured using a corneometer. Skin elasticity and firmness are measured using a cutometer. Wrinkle depth and density are measured using a profilometer or 3D imaging system. Skin redness and irritation are assessed using a chromameter or by visual scoring. Subject self-assessment questionnaires are used to evaluate perceived improvements in skin appearance and feel. Biopsies may be collected for histological analysis (collagen staining, elastin staining) and gene expression analysis. For anti-allergen studies, subjects with sensitive skin are exposed to UV radiation or chemical irritants (e.g., sodium lauryl sulfate) in the presence or absence of the peptide, and skin redness and irritation are measured.
