Product Description
Ordopidine is a novel and potent dopaminergic stabilizer
Ordopidine (CAS#: 871351-60-9), also known as ACR325, is a small molecule dopaminergic stabilizer developed for the potential treatment of Parkinson's disease and psychosis. With a molecular formula of C14H20FNO2S and a molecular weight of 285.38 g/mol, its IUPAC name is 1-ethyl-4-(2-fluoro-3-(methylsulfonyl)phenyl)piperidine. Ordopidine is a dopamine D2 receptor antagonist. It can inhibit hyperactivity caused by psychostimulants and stimulate behavior in states of hypoactivity. The compound significantly increases the level of dopamine and noradrenalin in the forebrain and concurrently inhibits the over-activity. Ordopidine has potential applications in treating a range of neuropsychiatric disorders due to its dopaminergic stabilizing effects. In May 2016, Phase-I clinical trials in Parkinson's disease in Germany was discontinued. The compound reached Phase II clinical trials. Ordopidine is for research use only and is not for human therapeutic use.
Biological Activity I Assay Protocols (From Reference)
Targets
Dopamine D2 receptor. Ordopidine is a dopamine D2 receptor antagonist. Dopamine D2 receptors are G protein-coupled receptors that play a critical role in the regulation of motor function, reward, motivation, and cognition. By antagonizing D2 receptors, Ordopidine modulates dopaminergic signaling. The compound is a dopaminergic stabilizer that can inhibit hyperactivity caused by psychostimulants and stimulate behavior in states of hypoactivity. Ordopidine significantly increases the level of dopamine and noradrenalin in the forebrain and concurrently inhibits the over-activity. This dual action of stabilizing dopaminergic tone makes Ordopidine a potential therapeutic agent for Parkinson's disease and psychosis.
ln Vitro
Ordopidine is a dopamine D2 receptor antagonist that can inhibit hyperactivity caused by psychostimulants and stimulate behavior in states of hypoactivity. The compound significantly increases the level of dopamine and noradrenalin in the forebrain and concurrently inhibits the over-activity. Ordopidine has potential applications in treating a range of neuropsychiatric disorders due to its dopaminergic stabilizing effects. The compound's activity has been characterized in receptor binding and functional assays measuring D2 receptor antagonism. Ordopidine is a small molecule dopaminergic stabilizer developed for Parkinson's disease and psychosis.
ln Vivo
Ordopidine has been studied in vivo in animal models and clinical trials. In May 2016, Phase-I clinical trials in Parkinson's disease in Germany was discontinued. The compound reached Phase II clinical trials. Detailed pharmacokinetic and pharmacodynamic data are available from preclinical and clinical studies. Ordopidine is for research use only and is not for human therapeutic use.
Enzyme Assay
Dopamine D2 receptor binding assays are performed using membranes prepared from cells expressing recombinant D2 receptors or from striatal tissue. Radioligand binding studies use [³H]-spiperone or [³H]-raclopride as labeled ligands. Membrane preparations are incubated with varying concentrations of Ordopidine and a fixed concentration of radioligand in binding buffer (50 mM Tris-HCl pH 7.4, 120 mM NaCl, 5 mM KCl, 2 mM CaCl₂, 1 mM MgCl₂) for 60-120 minutes at room temperature. Non-specific binding is determined using excess haloperidol. Bound radioactivity is measured by scintillation counting after filtration through GF/B filters. IC50 and Ki values are calculated by non-linear regression. Functional assays measure D2 receptor-mediated inhibition of cAMP accumulation.
Cell Assay
Cellular D2 receptor antagonism is evaluated in cell lines expressing recombinant D2 receptors (e.g., HEK-293 cells). Cells are cultured in appropriate media at 37°C with 5% CO₂ and treated with Ordopidine at various concentrations. Functional assays measure receptor-mediated inhibition of cAMP accumulation (Gi-coupled receptor) in response to dopamine or other agonists. The compound's antagonist activity is assessed by its ability to inhibit dopamine-induced inhibition of cAMP. Cell viability is assessed using MTT or LDH assays. Each experiment includes known dopamine receptor antagonists (e.g., haloperidol) as positive controls and vehicle controls.
Animal Protocol
In vivo efficacy of Ordopidine was evaluated in animal models of Parkinson's disease and psychosis, and in clinical trials. The compound is administered orally or intraperitoneally at doses determined by preclinical studies. In animal models, behavioral assessments include tests of motor function and psychostimulant-induced hyperactivity. Dopamine and noradrenalin levels in the forebrain are measured by HPLC. In May 2016, Phase-I clinical trials in Parkinson's disease in Germany was discontinued. The compound reached Phase II clinical trials. Sample sizes in clinical trials typically range from 20-100 patients per group.
