ODN-2336 is a novel A-Class CpG ODN (oligodeoxynucleotides) acting as a potent TLR9 agonist with the potential to be used as adjuvant of vaccines. It can induce the production of IFN-α and up-regulate the expression of IP-10 mRNA and IL-18 mRNA. Synthetic oligodeoxynucleotides (ODN) containing unmethylated deoxycytosine-deoxyguanosine (CpG) motifs are very potent inducers of the innate immune system, mimicking the effects of bacterial DNA. CpG ODN are recognized by Toll-like receptor 9 (TLR9). Three classes of TLR9 agonists have been described: B-Class CpG ODN that induce strong B- and NK-cell activation and A-Class ODN that induce very high levels of IFN-alpha by plasmacytoid dendritic cells. The recently described C-Class ODN combine most efficiently properties of A- and B-Class ODN in that they induce strong B-cell activation comparable to B-Class ODN together with IFN-alpha secretion comparable to A-Class ODN.

The expression of f CD69 and IFN-α is upregulated by ODN 2336 (0.5 µM; 24 48 hours) [1]. After 4 hours, ODN 2336 (3 µg/ml) promotes unusual transcription of IP-10 mRNA in human PBMCs [1]. At the 16-hour mark, ODN 2336 (3 µg/ml; 2, 4, 16 hours) increases the expression of IL-18 mRNA [1]. In adult and cord blood plasma, ODN 2336 (50 μg/mL; 20 hours) plus IL-3 (10 ng/mL) promotes the production of IFN-α [2]. Human PBMCs treated with ODN 2336 (0.3125-5 µg/ml) for 24 hours produce more FNA1 in a dose-dependent manner [3].
