| Targets |
N 556 is a prodrug that is converted to disodium cromoglycate, the active moiety. Disodium cromoglycate acts as a mast cell stabilizer, preventing the release of histamine and other inflammatory mediators from mast cells. It is used for the treatment of allergic asthma and allergic rhinitis. The mechanism involves inhibition of calcium influx into mast cells, preventing degranulation and mediator release.

| ln Vitro |
N 556 itself has limited in vitro activity as it is a prodrug. The active metabolite, disodium cromoglycate, inhibits mast cell degranulation and mediator release in vitro. It stabilizes mast cells by inhibiting calcium influx and preventing the release of histamine, leukotrienes, and other inflammatory mediators. In vitro assays measure inhibition of mast cell activation and mediator release.

| ln Vivo |
Research was done on KY-556 (N-556)'s oral antiallergic properties. Compared to intravenous disodium cromoglycate (DSCG), N-556 (10-100 mg/kg, po) suppresses homologous passive cutaneous anaphylaxis (PCA) in rats for 48 hours in a dose-dependent manner. In actively sensitized rat lung segments, N-556 (20 and 100 mg/kg orally once daily for 20 days) seems to decrease histamine release. Rats treated with N-556 (100 mg/kg, p.o.) showed extended survival from anaphylaxis. Rats with experimental asthma show a considerable reduction in airway resistance when given N-556 orally at a dose of 100 mg/kg [1].
In vivo studies demonstrate that N 556 is an orally active prodrug of disodium cromoglycate effective against allergic diseases. Upon oral administration, N 556 is absorbed and converted to DSCG, which then exerts anti-allergic effects. The compound has been evaluated in animal models of allergy and asthma. Oral administration of N 556 provides systemic delivery of DSCG, overcoming the poor oral bioavailability of the parent drug. |
| Enzyme Assay |
For prodrug conversion assays, N 556 is incubated with plasma or tissue homogenates, and the rate of conversion to disodium cromoglycate is measured by HPLC or LC-MS. Enzyme kinetics are determined to identify the esterases responsible for prodrug activation.
|
| Cell Assay |
For mast cell stabilization assays, rat peritoneal mast cells or RBL-2H3 cells are sensitized with IgE and challenged with antigen in the presence or absence of N 556 or DSCG. Histamine release is measured by ELISA or fluorometric assay. Calcium influx is measured using fluorescent calcium indicators.
|
| Animal Protocol |
For in vivo anti-allergic studies, rats or guinea pigs are sensitized with antigen and challenged to induce allergic responses. N 556 is administered orally at doses determined from pharmacokinetic studies. Endpoints include inhibition of passive cutaneous anaphylaxis, reduction in bronchoconstriction, and decreased inflammatory mediator levels in plasma or tissues.

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations
Oral Formulation 1: Suspend in 0.5% CMC Na (carboxymethylcellulose sodium) Oral Formulation 2: Suspend in 0.5% Carboxymethyl cellulose Example: Take the Oral Formulation 1 (Suspend in 0.5% CMC Na) as an example, if 100 mL of 2.5 mg/mL working solution is to be prepared, you can first prepare 0.5% CMC Na solution by measuring 0.5 g CMC Na and dissolve it in 100 mL ddH2O to obtain a clear solution; then add 250 mg of the product to 100 mL 0.5% CMC Na solution, to make the suspension solution (2.5 mg/mL, ready for use in animals). *Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
