Product Description
MLS1082 is a positive allosteric modulator (PAM) of the pyridine-based D1-like dopamine receptor with EC50 of 123 nM for dopamine-stimulated G-protein signaling.
MLS1082 is a positive allosteric modulator (PAM) of the D1 dopamine receptor. It potentiates dopamine-stimulated G-protein-mediated signaling and β-arrestin-mediated signaling with an EC50 of 123 nM. MLS1082 binds to a different allosteric site than MLS6585. It is a pyrimidone-based compound used as a chemical tool to study D1 dopamine receptor signaling and allosteric modulation.
Biological Activity I Assay Protocols (From Reference)
Targets
D1 dopamine receptor (D1-like dopamine receptor).
ln Vitro
MLS1082 improves D1R signaling mediated by G proteins and β-Arrestin. [1].
MLS1082 is a positive allosteric modulator of the D1 dopamine receptor that potentiates dopamine-stimulated G-protein-mediated signaling and β-arrestin-mediated signaling with an EC50 of 123 nM. It binds to a different allosteric site than MLS6585. The compound is a pyrimidone-based D1-like dopamine receptor positive allosteric modulator. MLS1082 is used as a chemical tool to study D1 dopamine receptor signaling and allosteric modulation.
ln Vivo
In vivo efficacy data for MLS1082 is limited. The compound is primarily used as an in vitro tool for studying D1 dopamine receptor signaling and allosteric modulation. As a positive allosteric modulator, it has potential applications in studying dopamine receptor function in neurological and psychiatric disorders. Further in vivo studies in animal models of dopamine-related disorders would be needed to assess its therapeutic potential.
Enzyme Assay
The in vitro receptor binding assay for D1 dopamine receptor allosteric modulation uses membrane preparations from cells expressing recombinant human D1 dopamine receptor. MLS1082 is incubated with the receptor preparation and a radiolabeled D1 receptor ligand (e.g., [3H]SCH23390) at varying concentrations (typically 0.1 nM to 100 µM). Allosteric modulation is assessed by measuring the effect of MLS1082 on dopamine binding affinity or by measuring dopamine-stimulated signaling in the presence of the compound. The EC50 for potentiation of dopamine signaling is calculated from dose-response curves.
Cell Assay
In vitro cellular assays are performed using cells expressing recombinant human D1 dopamine receptor (e.g., HEK293 cells). Cells are treated with MLS1082 at concentrations ranging from 0.01 to 100 µM in the presence of varying concentrations of dopamine. G-protein-mediated signaling is assessed by measuring cAMP accumulation using a competitive immunoassay (e.g., ELISA or HTRF). β-arrestin recruitment is measured using BRET or ELISA-based assays. The EC50 for potentiation of dopamine-stimulated signaling is calculated from dose-response curves.
Animal Protocol
In vivo animal model data for MLS1082 is not extensively reported in the literature. As a research tool for studying D1 dopamine receptor signaling, the compound would typically be evaluated in rodent models of dopamine-related disorders such as Parkinson's disease, schizophrenia, or addiction. Standard efficacy studies would involve compound administration (oral or intraperitoneal) followed by assessment of behavioral endpoints, motor function, and dopamine-related signaling markers. Pharmacokinetic studies would assess oral bioavailability, plasma half-life, and brain penetration.
ADME/Pharmacokinetics
MLS1082 has a molecular weight of approximately 400 g/mol and is a pyrimidone-based compound. It is soluble in DMSO and should be stored as a powder at -20°C. Pharmacokinetic parameters such as oral bioavailability, plasma half-life, and brain penetration are not extensively characterized in publicly available literature. As a research compound, it is typically used in in vitro assays at concentrations ranging from nanomolar to low micromolar.
Toxicity/Toxicokinetics
No detailed toxicity data is publicly available for MLS1082. As a research compound, it is not intended for human use and has not undergone formal toxicological evaluation. Standard preclinical safety assessments for dopamine receptor modulators would typically include selectivity profiling against a panel of dopamine receptor subtypes and related GPCRs, hERG channel inhibition for cardiac safety, and CYP450 enzyme inhibition for drug-drug interaction potential. The compound is supplied as a solid and should be stored desiccated at -20°C.
