Product Description
Miramistin is a drug used in Russia and Ukraine for treatment of genitourinary tract infections as an antiseptic or individual prophylaxis of venereal diseases.
Miramistin (CAS#: 15809-19-5) is a cationic antiseptic and disinfectant with broad-spectrum antimicrobial activity. It targets bacterial cell membranes through its cationic properties, binding to negatively charged bacterial surfaces, disrupting membrane integrity, and inducing cell death. Miramistin is also a potent inhibitor of human 5α-reductase type 1 (5α-R1) with an IC50 of 4.28 μM, 7.2-fold stronger than rat enzyme, suggesting additional pharmacological applications.
Biological Activity I Assay Protocols (From Reference)
Targets
The primary molecular target of Miramistin is the bacterial cell membrane. By utilizing its cationic properties, Miramistin binds to the negatively charged surface of bacteria, disrupting membrane integrity, inhibiting bacterial metabolism and growth, and inducing cell death. It also inhibits human 5α-reductase type 1 (5α-R1) with an IC50 of 4.28 μM, showing mixed inhibition through NADPH site binding confirmed by SPR.
ln Vitro
In vitro, Miramistin demonstrates broad-spectrum antimicrobial activity against gram-positive and gram-negative bacteria, fungi, and viruses. It disrupts bacterial cell membranes and inhibits bacterial metabolism. Additionally, it inhibits human 5α-R1 with an IC50 of 4.28 μM, 7.2-fold stronger than the rat enzyme, and shows mixed inhibition confirmed by SPR binding analysis.
ln Vivo
In vivo, Miramistin is used as a topical antiseptic and disinfectant for wound care, surgical scrubs, and environmental disinfection. It is not used systemically due to toxicity concerns. Its 5α-R1 inhibition activity suggests potential applications in androgen-related conditions, though this requires further investigation.
Enzyme Assay
For antimicrobial susceptibility testing, bacterial or fungal cultures are grown in appropriate media, and Miramistin is added at various concentrations. Growth inhibition is assessed by broth microdilution. MIC values are determined as the lowest concentration inhibiting visible growth.
Cell Assay
For 5α-reductase inhibition assays, human or rat 5α-R1 microsomes are incubated with Miramistin and NADPH. The substrate testosterone is added, and the conversion to dihydrotestosterone is measured by HPLC or LC-MS/MS. SPR analysis is performed to confirm NADPH site binding, and inhibition kinetics are analyzed by Lineweaver-Burk plots.
Animal Protocol
For in vivo studies, animal models of skin infection or wound infection receive topical Miramistin. Bacterial burden is quantified by colony counting. For 5α-R1 inhibition studies, rats receive Miramistin by systemic administration, and dihydrotestosterone levels in target tissues are measured by LC-MS/MS.

Note: Please be aware that the above formulations are for reference only. InvivoChem strongly recommends customers to read literature methods/protocols carefully before determining which formulation you should use for in vivo studies, as different compounds have different solubility properties and have to be formulated differently.

(Please use freshly prepared in vivo formulations for optimal results.)
Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 2.2772 mL 11.3861 mL 22.7723 mL
5 mM 0.4554 mL 2.2772 mL 4.5545 mL
10 mM 0.2277 mL 1.1386 mL 2.2772 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
