The primary molecular targets of Methiothepin maleate are serotonin receptors, particularly 5-HT2 and 5-HT1 subtypes. It acts as a potent antagonist at these receptors. The compound also has activity at dopaminergic receptors. By blocking serotonin receptors, Methiothepin maleate modulates serotonergic neurotransmission in the central nervous system.

| ln Vitro |
Methiothepinmaleate is an antagonist of the 5-HT receptor, exhibiting pKd values of 7.10, 7.28, 7.56, and 6.99 for 5-HT1A, 5HT1B, 5HT1C, and 5HT1D, respectively [1]. Thiotipine mesylate's pKd for 5-HT5A, 5-HT5B, 5-HT6, and 5-HT7 are, in that order, 7.0, 7.8, 8.74, and 8.99 [2]. Thiotipine binds with great affinity to 5-HT2A, 5HT2B, and 5HT2C, exhibiting respective pK values of 8.50, 8.68, and 8.35 [3].
In vitro, Methiothepin maleate binds with high affinity to 5-HT2 receptors and demonstrates antagonist activity in functional assays. It also binds to 5-HT1 receptor subtypes and differentiates 5-HT1D subtypes. The compound is a potent serotonergic antagonist used for pharmacological characterization of serotonin receptors. |
| ln Vivo |
In vivo, Methiothepin maleate produces antipsychotic effects in animal models. It modulates serum prolactin and growth hormone levels in humans. The compound acts as a serotonin receptor antagonist in the central nervous system, affecting dopaminergic and serotonergic neurotransmission.
|
| Enzyme Assay |
For serotonin receptor binding assays, membranes from cells expressing recombinant 5-HT2 or 5-HT1 receptors or rat brain cortex are incubated with subtype-selective radioligands and increasing concentrations of Methiothepin maleate. Nonspecific binding is determined with excess unlabeled ligand. Bound radioactivity is quantified by filtration and scintillation counting.
|
| Cell Assay |
For functional assays, cells expressing 5-HT2 receptors are stimulated with serotonin in the presence or absence of Methiothepin maleate. Calcium mobilization or inositol phosphate production is measured to assess antagonism. pA2 or IC50 values are calculated from dose-response curves.
|
| Animal Protocol |
For in vivo studies, rats or mice receive Methiothepin maleate by intraperitoneal or oral administration. Antipsychotic activity is assessed by behavioral tests. Serum prolactin and growth hormone levels are measured by radioimmunoassay. Dopaminergic and serotonergic system effects are evaluated by neurochemical measurements in brain tissue.

| Additional Infomation |
Methiothepin maleate is a maleate salt prepared by reacting Methiothepin with an equivalent amount of maleic acid. It is a potent 5-HT2 receptor antagonist and also possesses 5-HT1 receptor antagonistic activity. It can differentiate between the 5-HT1D subtype. Furthermore, it has affinity for rodent 5-HT5B, 5-HT5A, 5-HT7, and 5-HT6 receptors (pK1 values of 6.6, 7.0, 8.4, and 8.7, respectively). It can be used as a serotonergic antagonist, dopaminergic antagonist, and antipsychotic. It contains a Methiothepin (2+) group.
A serotonergic receptor antagonist in the central nervous system, used as an antipsychotic. |

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals).
