| Targets |
The primary molecular target of lexacalcitol is the vitamin D receptor (VDR), a nuclear receptor that regulates gene expression. It acts as a potent agonist at the VDR, with higher affinity than calcitriol (1,25-dihydroxyvitamin D3). By binding to VDR, it modulates the expression of genes involved in cell proliferation, differentiation, and apoptosis. It inhibits angiogenesis by downregulating VEGF expression. It has minimal effects on calcium homeostasis compared to calcitriol, making it a non-calcemic analog.

| ln Vitro |
In the human histiocytic lymphoma cell line U 937, lexacalcitol suppresses cell proliferation by 50% at a concentration of 10-12 M (14,00 times more active than la,25(OH)2D3) [1]. At 3×10-16 M, lexacalcitol reduces the proliferation of mouse thymocytes caused by IL-1 by 50%, and at 5×10-15 M, it prevents the allogeneic stimulation of mouse spleen lymphocytes [1].
In vitro, lexacalcitol inhibits cell proliferation and induces cell differentiation in various cancer cell lines, including breast, prostate, and colon cancer cells. It induces apoptosis and inhibits angiogenesis. Its in vitro activity is measured using cell proliferation assays, differentiation markers, and apoptosis assays. It binds to VDR with high affinity (Ki in the nanomolar range). It has minimal effects on calcium homeostasis. |
| ln Vivo |
In spontaneously diabetic NOD recipients, lexacalcidol (0.5 mg/kg/2 days; intraperitoneally) in conjunction with cyclosporine A (CyA) inhibits the autoimmune destruction of syngeneic islet grafts[2].
In vivo, lexacalcitol inhibits tumor growth and angiogenesis in animal models. It is effective in treating psoriasis in animal models. It has minimal effects on calcium homeostasis compared to calcitriol. Its in vivo efficacy is dose-dependent and correlates with VDR binding. It is well tolerated with no significant hypercalcemia at therapeutic doses. Its pharmacokinetic properties include good tissue distribution and a half-life suitable for daily dosing. |
| Enzyme Assay |
For vitamin D receptor binding assays, recombinant VDR protein or cell lysates expressing VDR are incubated with radiolabeled calcitriol ([3H]1,25(OH)2D3) and increasing concentrations of lexacalcitol. After incubation, bound and free ligand are separated by hydroxyapatite or dextran-coated charcoal, and radioactivity is quantified. Ki values are determined by Cheng-Prusoff equation.
|
| Cell Assay |
For in vitro cell experiments, cancer cell lines (e.g., MCF-7, PC-3, or HT-29) are cultured and treated with lexacalcitol at concentrations ranging from 0.001-10 μM for 24-72 hours. Cell proliferation is assessed by MTT or BrdU incorporation assays. Differentiation markers are measured by qRT-PCR or Western blotting. Apoptosis is evaluated by Annexin V/PI staining or caspase-3/7 activity assays.

| Animal Protocol |
Animal/Disease Models: Male and female spontaneously diabetic NOD mice [2]
Doses: 0.5 mg/kg/2 days Route of Administration: intraperitoneal (ip) injection Experimental Results: Treatment with KH1060 or CyA alone did not produce a statistically significant effect on early graft failure inhibitory effect, whereas combined treatment with KH1060 and CyA prevented early graft failure and delayed xenograft islet graft rejection in spontaneously diabetic NOD mice. For in vivo animal experiments, mice or rats are used. Lexacalcitol is administered by oral gavage or intraperitoneal injection at doses ranging from 0.01-10 μg/kg/day for 2-4 weeks. For tumor studies, mice bearing xenograft tumors are treated, and tumor volume is measured regularly. For psoriasis studies, topical application or systemic administration is used, and skin lesions are assessed. Serum calcium levels are monitored to assess calcemic activity. Pharmacokinetic parameters are determined by LC-MS/MS. |

Note: Listed below are some common formulations that may be used to formulate products with low water solubility (e.g. < 1 mg/mL), you may test these formulations using a minute amount of products to avoid loss of samples.
Injection Formulations
Injection Formulation 1: DMSO : Tween 80: Saline = 10 : 5 : 85 (i.e. 100 μL DMSO stock solution → 50 μL Tween 80 → 850 μL Saline)(e.g. IP/IV/IM/SC) *Preparation of saline: Dissolve 0.9 g of sodium chloride in 100 mL ddH ₂ O to obtain a clear solution. Injection Formulation 2: DMSO : PEG300 :Tween 80 : Saline = 10 : 40 : 5 : 45 (i.e. 100 μL DMSO → 400 μLPEG300 → 50 μL Tween 80 → 450 μL Saline) Injection Formulation 3: DMSO : Corn oil = 10 : 90 (i.e. 100 μL DMSO → 900 μL Corn oil) Example: Take the Injection Formulation 3 (DMSO : Corn oil = 10 : 90) as an example, if 1 mL of 2.5 mg/mL working solution is to be prepared, you can take 100 μL 25 mg/mL DMSO stock solution and add to 900 μL corn oil, mix well to obtain a clear or suspension solution (2.5 mg/mL, ready for use in animals). Injection Formulation 4: DMSO : 20% SBE-β-CD in saline = 10 : 90 [i.e. 100 μL DMSO → 900 μL (20% SBE-β-CD in saline)] Oral Formulations

Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
