Product Description
Fmoc-Cys(Trt)-OH-1,2,3-13C3,15N is a 15N-labeled and 13C (carbon 13)-labeled Fmoc-Phe-OH.
Fmoc-Cys(Trt)-OH-1,2,3-13C3,15N is a carbon-13 and nitrogen-15 labeled cysteine derivative with Fmoc and trityl protecting groups. The 13C3/15N label spans the cysteine backbone (C1, C2, C3 and N). It is used in solid-phase peptide synthesis and as a tracer for cysteine redox metabolism and protein turnover.
Biological Activity I Assay Protocols (From Reference)
Targets
No classical pharmacological target. Its functional role is as a labeled cysteine building block; the 13C3/15N label enables mass-spectrometric quantification of cysteine-containing peptides and tracing of cysteine incorporation into proteins and glutathione.
ln Vitro
Drug compounds have included stable heavy isotopes of carbon, hydrogen, and other elements, mostly as quantitative tracers while the drugs were being developed. Because deuteration may affect a drug's pharmacokinetics and metabolic properties, it is a cause for concern [75].
No IC50 or EC50 applies. Quality attributes include isotopic enrichment (≥98 atom% 13C and ≥98 atom% 15N), chemical purity, and coupling efficiency. These parameters ensure reliable performance as an internal standard or labeled precursor.
ln Vivo
No intrinsic in vivo pharmacological activity. In tracer studies, labeled cysteine is incorporated into proteins and glutathione, and the label is quantified to assess redox metabolism and protein synthesis.
Enzyme Assay
Purity and isotopic enrichment are verified by HPLC, LC-MS, and NMR. The compound is used to prepare internal standard solutions for quantitative cysteine and peptide analysis. High-resolution mass spectrometry confirms the 13C3/15N isotopic pattern.
Cell Assay
In cell-based studies, labeled cysteine is supplied in culture medium, and label incorporation into proteins or glutathione is measured by LC-MS/MS to assess redox flux and synthesis rates.
Animal Protocol
In vivo tracer studies administer labeled cysteine by infusion or feeding. Tissue proteins and glutathione are analyzed by mass spectrometry to determine 13C and 15N enrichment and calculate fractional synthesis rates.

Note: Please be aware that the above formulations are for reference only. InvivoChem strongly recommends customers to read literature methods/protocols carefully before determining which formulation you should use for in vivo studies, as different compounds have different solubility properties and have to be formulated differently.

(Please use freshly prepared in vivo formulations for optimal results.)
Preparing Stock Solutions 1 mg 5 mg 10 mg
1 mM 1.6958 mL 8.4792 mL 16.9584 mL
5 mM 0.3392 mL 1.6958 mL 3.3917 mL
10 mM 0.1696 mL 0.8479 mL 1.6958 mL

*Note: Please select an appropriate solvent for the preparation of stock solution based on your experiment needs. For most products, DMSO can be used for preparing stock solutions (e.g. 5 mM, 10 mM, or 20 mM concentration); some products with high aqueous solubility may be dissolved in water directly. Solubility information is available at the above Solubility Data section. Once the stock solution is prepared, aliquot it to routine usage volumes and store at -20°C or -80°C. Avoid repeated freeze and thaw cycles.
