NBP has proven to be an ideal solvent to replace the hazardous solvent DMF in solid-phase peptide synthesis (SPPS). This paper investigates the introduction of Fmoc-Arg(Pbf)-OH (one of the most difficult amino acids to handle) into growing peptide chains. The poor performance of this amino acid stems from its formation of a completely inactive δ-lactam, leading to reduced yields and often accompanied by the formation of the corresponding dearginine peptide. This problem is exacerbated when using NBP as a solvent, likely due to its high viscosity hindering the penetration of the coupling mixture into the resin. To address this issue, we propose a strategy for the safe introduction of Fmoc-Arg(Pbf)-OH into SPPS at 45 °C with minimal excess reagent: 1.75 equivalents of the protected amino acid, 1.8 equivalents of DIC, and 1.5 equivalents of OxymaPure. The core of this strategy is in-situ activation. Specifically, Fmoc-Arg(Pbf)-OH and OxymaPure dissolved in NBP were added to the peptide resin, heated to 45°C, and then half of the DIC was added and allowed to stand for 30 minutes. Then the other half of the DIC and some additional Fmoc-Arg(Pbf)-OH were added. Throughout the process, the temperature was maintained at 45°C. This serves two purposes: first, to reduce the viscosity of NBP, thereby promoting the penetration of the coupling mixture into the resin; and second, to accelerate the coupling reaction. It is expected that this strategy can be widely used to improve the performance of solid-phase peptide synthesis (SPPS), including the industrial preparation of peptides using this method. [1]
